| Wheat(Triticum aestivum)is one of the most important food crops in the world.The production of wheat is related to the stable development of the national economy and the health of people’s lives.Sitobion avenae,a common pest on wheat,has caused huge threats and losses to wheat production.Breeding and planting S.avenae-resistant varieties is one of the most important ways to solve this problem.In order to find out the resistance of 10 wheat cultivars to S.avenaes,this study measured the growth and development characteristics and feeding behavior of S.avenaes on wheat plants,and analyzed the physical,nutrient and secondary metabolic compounds of different wheat cultivars against S.avenaes.Metabolic basis,in order to lay a foundation for breeding excellent aphidresistant wheat varieties(lines).The main results obtained are as follows:1.The life parameters and feeding behavior of S.avenaes on the tested 10 wheat plants were clarified.Found through indoor wheat seedling infestation experiments,the developmental duration of S.avenaes in Mianyang 86-11,Baiyoumai,Granja de Melill and Kendih Aria no80 was significantly higher than that in 3 varieties of Jiyumai,Liuzhutou and Rivadeneira;The average daily number of eggs laid on the wheat plants of Jiyumai,Liuzhutou,Rivadeneira was significantly higher than that of the 4 varieties including Mianyang 86-11,Baiyoumai,Kendih Aria no80 and Rivadeneira;the average daily weight growth rate of S.avenaes on the two wheat varieties,the crucian carp wheat and BH941,was significantly higher In Mianyang 86-11.Using electrical penetration graph(EPG)to analyze the feeding behavior of Aphids in different varieties of plantlets,it was found that,compared with other species,the np,C,pd of S.avenaes feeding on Granja de Melill plants have longer duration;The occurrence frequency of probing behavior(pd)on Granja de Melill and Mianyang 86-11 plants was significantly higher than that on stigma cultivars.It showed strong epidermal resistance to S.avenaes;the E1 lasted longer,and the E2 was shorter,showing strong phloem resistance.There was no significant difference in G among the 10 wheat species.The 10 wheat varieties tested were grouped according to the data of the above 14 experimental results,among which Mianyang 86-11 and Granja de Melill were classified as aphid-resistant varieties;Shanyou 225,Shijiazhuang 54,Baiyoumai,Kendih Aria no80 and Rivadeneira were medium-resistant varieties;Jiyumai,Liuzhutou and BH-941 are susceptible species.2.Revealed the physical,nutritional and secondary metabolite basis of different resistance to S.avenae in 10 wheat varieties tested.The results of the determination of the epidermal hair of wheat plants by scanning electron microscope showed that there were differences in the density of the epidermal hair of different varieties of wheat.The densities of epidermal hairs of four wheat species,including Jiyumai,Liuzhutou,BH941 and Granja de Melill,were significantly lower than other varieties.Through the analysis of the nutrient components of wheat leaves by Gas Chromatography-Mass Spectrometer,it was found that there were differences in the nutrient components of different varieties of wheat leaves.The relative content of alanine in leaves of Mianyang 86-11 seedlings was significantly higher than that of other varieties.The relative content of tyrosine in the leaves of Granja de Melill seedlings was significantly higher than that of Shanyou 225;the relative content of tryptophan in the leaves of crucian carp and wheat seedlings was significantly lower than that of other varieties except Shijiazhuang 54;the relative content of aspartic acid in the leaves of Shanyou 225 seedlings Significantly higher than Jiyumai,Baiyoumai and Liuzhutou.It is speculated that the relative contents of alanine,tyrosine,tryptophan and aspartic acid may be related to aphid resistance in wheat.The relative content of glucose in seedling Jiyumai was significantly higher than that of other varieties,and the relative content of fructose was significantly lower than that of Shanyou225 and Shijiazhuang 54;It is speculated that the differences in resistance exhibited by Jiyumai and Shanyou225 may be related to the contents of fructose,glucose and sucrose in leaves.The metabolome of wheat plants was analyzed by Liquid Chromatography-Mass Spectrometer,and a total of 584 wheat metabolites were isolated and identified.There were significant differences in the metabolites of different varieties of wheat plants.Different metabolites in leaves of BH941 were screened out,and 34 metabolites that might be related to plant resistance were screened out.It is speculated that these metabolites may be associated with wheat-induced defense.In this study,two aphid-resistant wheat varieties and three aphid-susceptible wheat varieties were screened out by comparing the feeding behavior and life parameters of Aphids on 10 wheat plants.And by measuring the leaf epidermal hair density,the relative content of free amino acids and soluble sugar in the leaves,and the changes in the relative content of metabolites in the leaves before and after the induction of feeding by S.avenae.The physical,nutritional metabolism and secondary metabolite basis of different resistance of aphids can provide reference for the screening and breeding of wheat aphid resistance. |
