China is a big country of Chinese herbal medicine cultivation.With the expansion of Chinese herbal medicine market,the development of Chinese herbal medicine and Chinese patent medicine is more and more popular and recognized.However,the yield and quality of Chinese herbal medicine has not been improved and improved,the main reason is that soil-borne diseases can not be effectively controlled,and a series of quality problems caused by excessive pesticide residues have hindered the development of Chinese herbal medicine industry.This paper firstly introduces the current status and harm of pesticide use,the severity of soil-borne diseases of Chinese herbal medicine,and the advantages and disadvantages of three aspects of prevention and control measures.Then,the mechanism and advantages of Bacillus subtilis and the shortcomings of single agent are proposed,and the solution of compound bacteria agent is proposed to reduce the dosage and synergize.The proportion of principal components was determined by single factor test and orthogonal test was used to screen and optimize the excipients.And the quality standard evaluation,stability evaluation,antibacterial performance comparison evaluation.The type and concentration of peroxide and Bacillus subtilis were determined by single factor experiment,that is,the toxicity test of peroxide,the bacteriostatic test of Bacillus subtilis single agent and the compatibility test,and the main components were screened out by compounding different volume ratios.The optimal synergistic volume ratio.After determining the main components,orthogonal tests were carried out to screen the auxiliary materials,and the optimal formula of the compound bacterial agent was obtained:18%sodium peroxysulfate,18%Bacillus subtilis QST-713,20%potassium dihydrogen phosphate,20%dipotassium hydrogen phosphate,Manganese sulfate 12%,anhydrous magnesium sulfate 12%,and design reasonable usage and dosage.Secondly,the quality standard evaluation of the compound inoculum was carried out,including the quality evaluation of the Bacillus subtilis inoculum and the validation of the active oxygen method.The quality of the inoculum is evaluated by the appearance,the number of effective viable bacteria,the rate of miscellaneous bacteria,the moisture content,and the storage stability.The detection of active oxygen content mainly uses the iodometric method for methodological verification,and the linear regression equation is obtained as Y=4.383X+0.0532,(R~2=0.9997),the recovery rate is between 99.5%and101.5%,and the precision is high.The specificity is good,and the relative standard deviation is within 2%,which meets the requirements.Then,the stability of the Bacillus subtilis compound inoculum was evaluated:through the influence factor test and accelerated test,it was found that this product is most sensitive to high temperature and high humidity environment,and attention should be paid to heat insulation and moisture insulation during storage and transportation.The outer packaging is made of aluminum foil bags;the results of the long-term test tentatively set the validity period of the compound inoculum to be 12 months.However,in order to obtain an accurate validity period,it is still necessary to keep samples for observation and complete the 24-month and 36-month testing of this product.Finally,a comparative evaluation of the performance of the composite inoculants was carried out.Through the phosphorus-dissolving circle test and the soluble phosphorus content detection,the composite inoculant and the commercial samples were compared and evaluated,and the phosphorus-dissolved amount of the composite inoculant can reach352 mg/L,which can dissolve mineral phosphorus in the soil.The antibacterial properties of the Bacillus subtilis compound inoculation were compared and evaluated,and the commercially available samples were taken and the compound inoculation was tested for bacteriostasis.The results of the inhibition zone confirmed that this product can effectively inhibit Staphylococcus aureus ATCC6538,Escherichia coli 8099 and oxysporum Fusarium HG-11 can control bacterial soil-borne diseases,and its bacteriostatic effect is better than that of commercial samples.It is of great significance for the prevention and control of soil-borne diseases of Chinese herbal medicine and the most fundamental drug safety.
