Dendrobium officinale Kimura et Migo,as "gold in medicine",has a high medicinal value.However,due to excessive mining and waste of medicinal byproducts,the wild resources of D.officinale are increasingly scarce.More and more researchers have applied plant tissue culture and human culture technology to the production of Medicinal D.officinale.As a typical symbiotic plant,D.officinale has been greatly influenced by symbiotic fungi in its growth and the quality of active products.The study on the symbiosis mechanism between D.officinale and fungi is an important part of the artificial breeding of D.officinale.However,previous studies on the cover of different fungi are not comprehensive,and few studies on molecular level.The study of long noncoding RNA(lnc RNA)in D.officinale has not been found.In this experiment,three kinds of fungi were extracted from D.officinale,respectively Fusarium sp.,Mycena sp.and Tulannella sp.After single and mixed symbiosis culture with D.officinale,the physiological indexes,stem polysaccharides and leaf flavonoids content of D.officinale were determined.Compared with sterile seedlings,the plant height and root length of fungal seedlings grew significantly,while sterile seedlings showed yellowing of leaves and decrease in fresh weight at the later stage of culture.The stem polysaccharide and leaf flavonoid contents of the fungal seedlings were higher than those of the sterile seedlings.The effects of different genera on the growth and development of D.officinale showed differences.The most important factor to promote the growth of fresh weight and plant height of D.officinale was the Tulasnella sp.,the largest promoting effect on root length growth was Mycena sp.and Fusarium sp was the most important to dry weight accumulation.The difference of the different fungi on the accumulation of active substances in D.officinale was also different,among which the most influential was the Tulasnella sp.,and the less affected were Fusarium sp.and Mycena sp.,which was the most significant in the accumulation of stem polysaccharide content.The effect of mixed culture of three fungi was better than that of the single culture.In this experiment,26543 groups of lnc RNA were obtained by high-flux sequencing of the seedlings and sterile seedlings of D.officinale.The results showed that 1275 groups of go enrichment data and 20 KEGG enrichment pathways were obtained.Among them,10 groups of lnc RNA significantly up regulated in the vaccine and 10 groups of significantly down regulated lnc RNA were selected to predict the regulatory relationship between lnc RNA and m RNA by bioinformatics.The selected lnc RNA involved five enrichment KEGG pathways,which were up regulated in Starch and sucrose metabolism and Phenopropanid biosynthesis,down regulated in Plant hormone signal transduction and Protein processing in endoplasmic reticulum.while in the Splicing pathway,there are target m RNA with up-regulated and down regulated expression.In the seedlings,the expression level of the related genes of sucrose synthetase 2 increased,which proved that symbiotic fungi promoted the accumulation and growth of polysaccharides in D.officinale by promoting sucrose metabolism;The up regulation of phenylpropenol dehydrogenase 4,a key enzyme in cell wall synthesis,proved that symbiotic fungi played a role in supporting and anti lodging of plants;The up regulation of UBP1 related protein gene proved that symbiotic fungi increased the level of host metabolism;The down regulation of RDM16 provides a new molecular level idea for the formation and regulation of D.officinale mycorrhiza;The down regulation of F-box protein GID2 and heat shock protein processed in endoplasmic reticulum in plant hormone signaling pathway indicates that symbiotic fungi play an important role in the anti reverse reaction of D.officinale.The molecular mechanism of fungal symbiosis affecting the growth and development level and active product content of D.officinale was studied in this experiment.It laid a foundation for the exploration of lnc RNA regulation network of D.officinale,which is helpful to the prevention of diseases and abnormalities of D.officinale.It has a certain application value in the field of artificial breeding of D.officinale. |
