Trichinella spiralis belongs to worms and is distributed all over the world.After infecting the host,it can cause trichinosis,which is a serious zoonotic parasitic disease.Previous studies have shown that infection with worms often leads to the aggregation of a large number of eosinophils in peripheral blood to resist the invasion of worms.However,it is unclear how eosinophils change from the time of eating undercooked meat containing larval cysts to the time when Trichinella spiralis forms cysts in the host muscle,and the mechanism of their increase has not been reported.Therefore,this subject has conducted a preliminary study on the dynamic changes and increasing mechanism of eosinophils in mice infected with Trichinella spiralis.Protein post-translational modification(PTMs)can affect the activation,survival,proliferation,differentiation and migration of immune cells,and are the key regulators of immune response.So after mice infected with Trichinella spiralis,is the change of eosinophils as immune cells related to the level of protein post-translational modification? Therefore,taking this as the starting point,this study conducted a preliminary study on eosinophils.Balb/c mice were infected with Trichinella spiralis muscle larvae by gavage,and the changes of eosinophils were detected by flow cytometry every day.The results showed that eosinophils showed a bimodal trend,and the proportion of eosinophils peaked on the 14 th and26th day after infection,especially on the 26 th day.Therefore,we selected eosinophils from mice infected with Trichinella spiralis on the 26 th day as the research samples,and detected Oglycosylation,acetylation,trimethylation,ubiquitination,monomethylation,crotonylation,dihydroxyisobutyrylation,lactation,malonylation,succinylation and phosphorylation by flow cytometry,The results showed that the phosphorylation level of eosinophils whole protein changed most significantly before and after infection.In view of the fact that the increase of eosinophils is related to JAK-STAT5 signaling pathway in the study of eosinophils model.Therefore,this subject detected the activation of JAK-STAT5 signal pathway.It was found that when mice were infected with Trichinella spiralis for 26 days,the phosphorylation level of STAT5 in eosinophils increased significantly.In order to further explore the regulatory factors related to the phosphorylation level of STAT5,we used a multi factor detection kit to detect the secretion levels of GM-CSF,IL-3,IL-5 and IL-6 in the serum of Trichinella spiralis infected mice.The results showed that the secretion level of IL-6in the serum decreased significantly.In conclusion,eosinophils in mice infected with Trichinella spiralis showed a bimodal trend.The proportion of eosinophils peaked on the 14 th and 26 th days after infection,especially on the 26 th day of infection;Therefore,the increase of eosinophils may be due to the significant decrease of IL-6 level in serum after infection with Trichinella spiralis,which leads to the significant increase of STAT5 phosphorylation level in peripheral blood eosinophils,and finally leads to the increase of peripheral blood eosinophils in mice infected with Trichinella spiralis.This study laid a foundation for further study on the mechanism of eosinophils in peripheral blood of infected Trichinella spiralis by using signal pathway inhibitors. |
