Enterotoxigenic Escherichia coli(ETEC)is an important pathogen causing diarrhea in human and young animals.ETEC adheres to the intestinal mucosa by the colonization factor(adhesin),which destroys various intestinal functions,and can indirectly lead to diarrhea,and diarrhea can cause the change of intestinal flora.Egg protein transferrin-deriver peptides IQW(Ile-Gln-Trp)and IRW(Ile-Arg-Trp)have been proved to have the effects of antihypertensive,antibacterial,anti-oxidation,anti-cancer,effective cardiovascular disease alleviation,endothelial inflammation and oxidative stress.This study explored the effects of IQW and IRW on intestinal histology injury,the content of inflammatory factors in tissue and intestinal microorganism in mice jejunum.In the ETEC induced model of jejunal inflammation,IQW and IRW regulated the mechanism of jejunal inflammation by alleviating the injury of jejunum,reducing inflammatory response and regulating the diversity and composition of intestinal microorganisms.Experiment 1: the effects of egg active peptides IQW and IRW on the tissue injury and inflammatory response of mice with jejunal inflammation caused by ETEC.48 ICR mice of 8 weeks old were randomly assigned to four groups after three accommodative days:(1)control group(CTRL): Mice were fed a normal diet and gavaged with saline.(2)ETEC group(ETEC): Mice were fed a normal diet and given ETEC by gavage;(3)IQW group(IQW+ETEC): Mice were fed a normal diet,fed water with IQW and gavaged with ETEC.(4)IRW group(IRW+ ETEC): Mice were fed a normal diet,fed water with IRW and gavaged with ETEC.The results showed that:(1)compared with the control group,the mice injected with ETEC intraperitoneally had serious histological injury,increased inflammatory cell infiltration and abnormal intestinal villi intestinal crypt.However,egg active peptide IQW and IRW could alleviate the jejunal tissue injury caused by ETEC,reduce the infiltration of inflammatory cells,and also make the intestinal villi and crypt behave normally.(2)The content of TNF-α,IL-1 β,IL-6 and IL-10 in jejunum of mice was increased by the addition of ETEC,while the content of TNF-α,IL-1 β,IL-6 and IL-10 was significantly reduced by egg active peptide IQW(p<0.05);IRW could also significantly reduce the contents of TNF-α,IL-6 and IL-10(p<0.05).Experiment 2: the effect of egg active peptides IQW and IRW on intestinal microorganisms in mice with jejunal inflammation induced by ETECIn Experiment 1,the effects of egg active peptide IQW and IRW on intestinal microorganisms in mice with jejunal inflammation induced by ETEC were also studied.The results showed that:(1)the egg active peptides IQW and IRW can increase the diversity of intestinal microorganisms of mice,specifically increase the abundance of the intestinal content microbial community(Chao index and ACE index),Shannon index(p<0.05),and reduce Simpson index(p<0.05).IQW and IRW can increase the content of Bacteroides at the level of the Phylum(p<0.05).In the class of the Class,the content of Bacteroidia can be increased by IQW and IRW(p<0.05),IQW also reduced the proportion of Flavobacteriia(p<0.05).At the Order level,IQW and IRW could reduce the content of Flavobacterials(p<0.05).At the level of family,IQW can reduce the content of flavobacteriaee(p<0.05).in the level of Genus,IQW and IRW can increase the content of Bacteroides(p<0.05).(2)The content of Flavobacteriia was correlated with IL-10,pearson=0.416(p=0.043);the content of Flavobacteriales was correlated with IL-10,pearson=0.416(p=0.043);the content of Flavobacteriaceae was correlated with IL-10,pearson=0.416(p=0.043).In conclusion,under the ETEC induced jejunal inflammation model,egg active peptides IQW and IRW can alleviate the jejunal injury and reduce the content of inflammatory factors in mice,improve the composition of intestinal microorganisms and improve the diversity of intestinal microorganisms,thus maintaining the stability of intestinal flora.
