| In this study,the common pathogenic microorganisms in the aquaculture industry were taken as the research object.Based on the fluorescence-photoelectric recording microbial detection instrument FORBIDTM Microbial Rapid Analyzer independently developed by our research group,the accurate and efficient specific detection strategy of pathogenic microorganisms was explored,and two methods were constructed.The rapid detection mode of various pathogenic microorganisms provides new methods and ideas for the detection of pathogenic microorganisms.The main research contents are as follows:(1)The rapid detection strategy of Escherichia coli and Aerobic plate count was explored by using fluorescence and colorimetric detection principles respectively.The specificity,accuracy and sensitivity of the target strains were evaluated,and the detection standard curve of the target strains was established.The detection strategy was successfully applied to the detection of actual samples.The experimental results show that our detection strategy can achieve ultra-sensitive detection of target microorganisms with a span of 100-106 CFU/m L of 7 orders of magnitude,and it performs well in the mixed detection of Escherichia coli,Salmonella typhi,Pseudomonas aeruginosa,and Staphylococcus aureus.Excellent specificity was obtained,and good accuracy was still obtained when the strategy was applied to the detection of real samples.Compared with the traditional recognized plate counting method,the detection results of this strategy have higher accuracy and stability;and the operation process is simple,the detection time is short,and the detection cost is low.The successful realization of this strategy can provide new ideas for the detection of pathogenic microorganisms.(2)A new mode of simultaneous detection of Escherichia coli and total coliform based on FORBIDTM Microbial Rapid Analyzer was explored,and it was successfully applied to the detection of aquaculture water quality.Establish a standard curve through the target strain,conduct sensitivity,linear range analysis and accuracy test,and explore its accuracy,efficiency and universality in the detection of various types of actual samples.The established detection method can realize the simultaneous detection of Escherichia coli and total coliforms,and the detection sensitivity range spans 100-104 CFU/m L.The detection of 3 different contamination levels,8 different matrices and 5 field samples showed extremely high accuracy.The advantages of detection are:wide sensitivity range,high accuracy,no need to predict the degree of sample contamination in advance;short detection time,all detection can be completed within 20 hours;one-step fool-like operation,which reduces the difficulty of operation;two indicators simultaneously detection,so that the detection cost is low.This model opens up new ideas for the detection of pathogenic microorganisms in the aquaculture industry,and has high promotion and application value. |
