| Arbuscular mycorrhizal fungi(AMF)is a kind of beneficial fungal microorganism that can form a good symbiotic relationship with most land plants.The symbiosis between AM fungi and host plants can promote the absorption of nutrients and water and improve the stress resistance of plants.It is of great significance to develop AM fungicide fertilizer for crop growth and ecological environment protection because of soil acidification and compaction caused by long-term application of chemical fertilizer.Among them,AM fungal spore propagation and spore resistance storage activity are the key technologies for AM fungal fertilizer production.In this study,AM fungal spores were stored at 25℃,4℃,-20℃and-80℃with three protective agents,glycerin aqueous solution,glycerin,normal saline and trehalose solution,and at different storage temperatures.The spore activity,infection and symbiosis rate of AM fungi under different storage conditions and physiological indicators of host plants after symbiosis were studied,and the main results were as follows:1.Survival rate and viability analysis of AM fungal spores under different storage conditions showed that the protective effect was glycerin solution>glycerin and saline mixture>trehalose solution.At 4℃>-20℃>25℃>-80℃,the spore survival rate was only1.67% lower than that of the control when stored at 4℃and glycerin solution for 6months,and the spore viability was 42% higher when stored at 4℃and trehalose solution for 6 months.2.Analysis of infection rate and infection intensity of AM fungal spores under different storage conditions showed that the protective agent effect was trehalose solution>glycerin solution>glycerin and normal saline mixed solution,and the temperature effect was4℃>-20℃>25℃>-80℃.The infection symbiosis rate of AM fungal spores treated with trehalose solution at 4℃could reach 88%.The infection intensity was high and arbuscular development was good.3.AM fungal spores under different storage conditions infect lotaustralin root symbiotic50 days of analysis can be seen that the influence of pulse root growth,different storage time and storage conditions on the symbiosis plant roots,root and lateral root,soluble sugar content and chlorop Hyll content differences,4℃and trehalose solution storage 6 months spores can significantly promote the root growth of arteries and veins,This further supported that spores stored at 4℃and trehalose solution for 6 months had higher viability and infection symbiosis rate.In conclusion,studies under different storage conditions showed that spores stored at 4℃and in trehalose solution for 6 months still maintained high activity and infection symbiosis rate,which provided experimental basis for the production and application of AM fungal agent(fertilizer). |
