| Seahalpolysaccharide has anti-tumor,antioxidant,immunomodulatory and anti-cardiovascular activities.In this study,18 one-year-old healthy beagles were randomly divided into control group(basic diet + starch capsule),low dose group(0.2g / kg body weight)and high dose group(basic diet + seaweed polysaccharide capsule 0.8g / kg).Blood samples were collected at 0,7,14,1,21 and 28 d after 28 days.On the 28 th d,samples of spleen,duodenum,middle jejunum and ileum were dissected.Changes of antioxidant indexes,immunoglobulin and cytokines in dog serum by blood routine and ELISA,relative expression of My D88 and TRAF-6 protein by IHC,quantitative P g A,TLR4,TLR7,My D88,TRAF-6 and AP-1B1 and expression of My D88,TLR7 and TRAF-6 protein to explore the effect of different doses of seaweed polysaccharide on the immune function of dogs.The test results are as follows:(1)The routine blood results showed that the number of leukocytes and neutrophils was significantly higher in the blood at the 7th and 14 th d in the low-dose group compared with the control group.(2)The immunoglobulin results showed that Ig A levels in groups 7,14,21 and 28 d compared with the control group.(3)The results showed that CAT was significantly increased in the 14 d low dose group compared with the control group.(4)The results of fluorescence quantitative PCR showed that the relative expression of m RNA in the duodenum of TLR-4 and TRAF-6 in the low-dose group and TLR-4,TRAF-6 in the jejunum,My D88,TLR-7,TRAF-6,AP-1B1 in the duodenum,TLR-4,TLR-7,TRAF-6,AP-1B1 in the jejunum,My D88,TLR-4,TLR-4,TLR-7 in the ileum,The relative expression of m RNA of TRAF-6 and AP-1B1 was significantly increased.(5)Protein blotting results showed that the relative expression of TRAF-6 protein in duodenum,jejunum and ileum in lower-dose group compared with control group;My D88,TLR-7,TRAF-6,TRAF-7,T R D 88,T y D-8,TLR-7 and TRAF-6 in duodenum in high-dose group.The results showed that feeding seaweed polysaccharide increased by increasing the water of immunoglobulin and cytokines in canine serum Ping,antioxidative damage ability,intestinal secretory Ig A expression and Toll-like receptor signaling pathway related protein expression,to play its role in enhancing body immunity. |
