Inverted nipple disease of pigs is one of the most common reproductive defect diseases in pig breeding,which leads to lack of milk,seriously affects the growth and development of piglets and brings economic losses to farmers.In order to reduce the incidence of inverted nipple disease,this study selected Anhui local breeds Wannan Hua pig as the research object to screen individuals prone to inverted nipple disease.1.In this study,164 Wannan Hua pigs were randomly selected,of which 29 were nipple invagination samples and 135 were normal nipple samples.In order to study the association between LEF-1,RLN and PTHR1 gene polymorphisms and nipple invagination disease in pigs,the distribution of LEF-1,RLN and PTHR1 gene polymorphisms in nipple invagination samples was detected and the association analysis was carried out.The results showed that the C position at position 118806 of DNA of porcine LEF-1 gene was associated with inverted nipple disease,and individuals with CC genotype were more likely to develop inverted nipple than individuals with AA genotype.No diversity was found at Tmax G site at position 1320 of DNA of RLN gene,and there was no close association between Chand T site at position 25195 of DNA of PTHR1 gene and inverted nipple disease.The results showed that porcine nipple invagination disease was closely related to the genotype of Amax C site at position 118806 of LEF-1 gene DNA.2.In order to further study the relationship between the genotype of LEF-1 gene and nipple invagination disease,this study randomly selected 55 pigs from the new black pig population in Yixian as samples for LEF-1 Gene polymorphism detection,to study the application of LEF-1 gene polymorphism in pig nipple retraction disease.In this experiment,three genotypes of AA,AC,and CC were detected in the population.According to the detection results,individuals who are susceptible to nipple retraction disease can be effectively screened.The incidence of inverted nipple disease has practical significance.
