Fusarium verticillioides is the main causal agent of maize ear rot.It leads to the loss of maize yield and the production of fumonisins,and harmful to humans and animals.Epigenetics is a mature method to study the mechanism of fungal diseases.It has been reported that histone deacetylase(HDACs)is involved in the reproductive growth and toxin synthesis of filamentous fungi.However,HDACs has not been reported in Fusarium verticillioides.Therefore,this paper uses the basis of molecular biology to explore the role of FvHos2 gene and FvHos3 gene in Fusarium verticillioides.In this study,the mutantsΔFvHos2,ΔFvHos3,ΔFvHos2-C andΔFvHos3-C were obtained by double-joint PCR and gene complementarity,and the genes functions of FvHos2 and FvHos3 were identified by a series of basic biological phenotypic experiment-s.1.The deletion mutantsΔFvHos2 andΔFvHos3 are defective in vegetative growth and conidiation.2.Sensitivity measurement assays showed that the deletion mutantΔFvHos2 are susceptive to Fludionil,Pydiflumetofen,SDS,CR,H2O2,Na+,K+,Ca2+and Mg2+,while the deletion mutantΔFvHos3 was more sensitive to Fludionil and H2O2,more tolerant to SDS,Ca2+and Mg2+,with the same as the wild type 7600 to Pydiflumetofen,CR,Na+and K+.3.With the virulence and pathgenicity,the deletion mutantsΔFvHos2 andΔFvHos3are defective.4.The deletion mutantsΔFvHos2 andΔFvHos3 more sensitive to the histone deacetylase inhibitor.5.Finally,the Panobinostat was selected to further analyze the genes function of FvHos2 and FvHos3.Panobinostat weakens the inhibitory effect of the deletion mutantsΔFvHos2 andΔFvHos3 in vegetative growth,conidiation,FB1 production and pathogenicity.To sum up,FvHos2 and FvHos3 genes may be involved in regulating the growth,the virulence and pathogenicity of Fusarium verticillioides.
