Semen cryopreservation is of great value in livestock breeding and breed preservation.The freezing resistance of sperm varies according to breeds and individuals,and the disc overy of sperm freezing tolerance biomarkers or genes can help to select and breed seme n freezing tolerant breeds.In this study,we selected the semen plasma of Huoshou Black pigs with high cryotolerance and the semen plasma with low cryotolerance as materials,and detected and analyzed the composition differences of semen plasma metabolites bet ween two groups by metabolomic technology,in order to provide a basis for screening an d predicting the biomarkers related to cryotolerance in the semen plasma of Huoshou Bla ck pigs.The details of the study and the results are as follows.Semen from 15 Huoshou Black Boar breeders was frozen and thawed for testing.Base d on the ratio of post-thaw viability to fresh semen viability,three boars with the highest sperm cryotolerance were selected as one group,and three boars with the lowest sperm cr yotolerance were selected as another group,and semen plasma was collected from both g roups for metabolomic examnination.The results of metabolome sequencing and bioinfo rmatics analysis showed that there were 621 different chemical types of metabolites in th e semen plasma of Huoshou black pigs.There were significant differences in the expressi on of metabolites in the semen plasma between the high and low cryotolerance groups.F urther study revealed that there were 185 different metabolites and a total of 4902 differe ntially expressed metabolic ions,of which 1150 were up-regulated and 3752 were downregulated.The results of the pathway analysis showed that the different metabolites in Hu oshou Black pig semen plasma were enriched in cellular processes,environmental infor mation processing,gene information processing,disease,metabolism and biological syst em pathways,mainly involving the metabolism of amino acids,carbohydrates and lipids.In summary,this paper presents the first metabolomic picture of Huoshou Black pig se men plasma,revealing the differences in the metabolomic profiles of high and low freezi ng tolerance boar semen plasma.Adenosine monophosphate,pyrogallocatechuic acid a,a nd n-acetyl dihydrosphingosine,palmitoylserine,and xanthine might be used as potential biomarkers of freezing tolerance in Huoshou Black pig sperm.The results of the study h ave important reference value for the freezing and preservation of Huoshou Black pig se men and even the optimization of germplasm conservation program.
