Aquilaria sinensis is a rare and endangered plant unique to China.It is a tree of Aquilaria of Thymelaeaceae.It is mainly distributed in the low-altitude areas of tropical and subtropical China.Aquilaria sinensis is evergreen all year round,with beautiful tree shape and excellent beautification effect.It has high landscape value.As a landscape tree or cultivated with other flowers,it has great landscape advantages.However,in recent years,the wild resources of Aquilaria sinensis have been seriously persecuted.There is an urgent need to protect the resources and excavate the genetic status of Aquilaria sinensis.So,it is of great significance to study the inheritance of Aquilaria sinensis resources.In this experiment,so149 families from 13 provenances such as Mashan,Guangxi,Pubei,Beiliu,Yunnan Pu’er,Tunchang,Danzhou,Qionghai,Wanning,Qiongzhong,Chengmai,Haikou,Ledong and Lingshui genetic and molecular marker analysis of physiological and biochemical indexes.Finally,after the distance degree and clustering graph are obtained,the corresponding genetic structure is analyzed.Then,the traits and molecular marker results obtained from the research and measurement are studied and analyzed,and finally the key relationship between a locus and traits is determined.Finally,the dominant provenances and classification under different conditions are obtained,which is helpful to explore the law of genetic variation It is also of great significance to reveal the spatial differences and genetic relationship between individuals and provenances,so as to provide a theoretical basis for the protection of existing Aquilaria sinensis resources.The main conclusions are as follows:(1)The phenotypic variation coefficient of Aquilaria sinensis was 1.415%-53.878%,with an average of 23.435%;The average of physiological traits was18.148%,and the variation range was 1.340% ~ 49.779%;Nei’s gene diversity index(H)ranged from 0.1719 to 0.2706,and the average value of population level was 0.3104.The phenotype,physiological characters and ISSR Molecular Markers of soil Aquilaria sinensis reached significant differences within and among provenances,indicating that there were significant genetic variations at provenances and individuals.(2)The(VST)was 1.715% ~ 99.554%,average of 65.700%;The variation range of physiological genetic different(VST)was 19.44% ~ 96.53%,and the mean value was 60.290%;The molecular marker GST is 0.7350.The comprehensive phenotypic traits,physiological traits and molecular marker molecules show that most of the variation of soil Aquilaria sinensis comes from between provenances and is relatively stable within provenances,which belongs to moderate genetic and environmental control.(3)Through comprehensive principal component analysis and cluster analysis,the 13 provenances of Aquilaria sinensis can be divided into three groups.The first is the provenance of Mashan,Pubei and Yunnan;The second type is Beiliu,Danzhou,Qiongzhong,Qionghai and Wanning provenances;The third category is Tunchang,Chengmai,Haikou,Ledong and Lingshui provenances.the same as those of molecular marker classification,indicating that gene is the core factor determining the inheritance and provenance classification of Aquilaria sinensis.(4)Comprehensive phenotypic,physiological and molecular genetic analysis,according to Mantel’s test,it shows that the geographical distance of provenance is significantly correlated with the genetic European distance,indicating that the genetic distance of Aquilaria sinensis can be explained by geographical distance.There are frequent gene exchanges between the provenances in Hainan and Guangxi.The provenances in Yunnan and Guangxi have high genetic diversity.It is the center of the genetic diversity of the provenances and can provide materials for parent selection.Hainan has the farthest genetic distance and is suitable for in-situ protection of germplasm resources.(5)Through the analysis of genetic structure by structure software,it can be divided into three subgroups.Based the general linear model,GLM detected 19 ISSR markers associated with soil Aquilaria sinensis traits,and a total of 16 traits could be associated with ISSR Molecular markers,in which the interpretation rate of trait variation ranged from 0.192% to 30.917%.The effect values of 16 soil Aquilaria sinensis traits associated with ISSR Molecular Markers ranged from-18.029 to 23.109,and the physiological effect value varied greatly compared with the phenotypic effect value.
