| In recent years,oxidative stress has become one of the hotspots in animal nutrition and health research.It seriously reduces the production performance and intestinal health of pigs.Oxidative stress is one of the important factors that reduce the antioxidant capacity and increase the content of ROS in pig intestine,which seriously reduces the production performance and intestinal health of pigs.Mitophagy is a process of selectively removing damaged and aging mitochondria through autophagy,which can reduce the release of reactive oxygen species into the cytoplasm.It is a delicate cellular self-protection mechanism.An important mechanism regulating cellular oxidative stress injury.E3 ubiquitin ligase(Parkinson’s disease gene 2,Parkin)is an important regulator of mitophagy,and Parkin-mediated mitophagy is the most important way to protect mitochondria from oxidative stress damage.Resveratrol(RES)is a plant polyphenol extracted from grapes,mulberries and other plants,and is a natural antioxidant.In this study,RES was added to the diet to feed finishing pigs,and the oxidative stress model of porcine intestinal epithelial IPEC-J2 cells was constructed by using Diquat(DIQ)to explore the effect of RES on activating Parkin-mediated mitophagy in pigs.The role and mechanism of intestinal antioxidant capacity provide a scientific theory for the future development of RES as a dietary additive that can relieve oxidative stress in pig intestinal tract and improve pig intestinal health by enhancing pig intestinal antioxidant capacity.In accordance with.The main findings of the test are as follows:1.The effect of adding RES in the diet on the antioxidant capacity and the expression of mitophagy-related proteins in the jejunum of finishing pigs.A total of 12 Duroc ×Landrace × Yorkshire grown pigs with an average body weight of about 65 kg were selected and randomly divided into a control group and an experimental group,with 6 pigs in each group.The control group was fed the basal diet,and the experimental group was supplemented with 400 mg/kg RES on the basal diet,the formal test cycle is 41 days,the weight is about 110 kg,the slaughter test is carried out,the fasting is 12 hours before slaughter,and the jejunal tissue is collected for storage.The test results show:(1)RES significantly decreased the content of MDA in pig jejunum,and significantly increased the activities of antioxidant enzymes T-SOD,CAT,GSH-Px and T-AOC in pig jejunum.(2)RES significantly increased ATP production in pig jejunum,activated Parkin-mediated mitophagy,up-regulated the expression of mitophagy-related proteins PINK1,Parkin,LC3II/LC3 I,and down-regulated the expression of p62,thereby increasing the key antioxidant enzyme genes in pig jejunum Expression of T-SOD,CAT and GSH-Px to improve the antioxidant capacity of pig intestine.2.The mechanism of RES improving the antioxidant capacity of porcine intestinal epithelial IPEC-J2 cells based on Parkin-mediated mitophagy.(1)DIQ significantly increased the content of ROS in IPEC-J2 cells,increased the content of MDA,and significantly decreased the activities of antioxidant enzymes T-SOD,CAT,GSH-Px and T-AOC,indicating that the oxidative stress model of IPEC-J2 was successfully established.(2)DIQ treatment of IPEC-J2 cells significantly decreased ATP production and mitochondrial membrane potential in IPEC-J2 cells,resulting in mitochondrial dysfunction;RES treatment significantly increased ATP production and mitochondrial membrane potential in IPEC-J2 cells under oxidative stress conditions,effectively alleviate DIQ-induced cellular mitochondrial dysfunction.(3)RES treatment of DIQ-induced oxidative stress cells can activate Parkin-mediated mitophagy,up-regulate the expression of mitophagy-related proteins PINK1,Parkin,LC3II/LC3 I,down-regulate the expression of p62,and significantly increase the antioxidant enzyme gene T-SOD the expression of,CAT and GSH-Px significantly increased the activities of antioxidant enzymes T-SOD,CAT,GSH-Px and T-AOC,indicating that RES can effectively relieve DIQ-induced oxidative stress in IPEC-J2 cells.(4)After the application of si RNA Parkin to inhibit the expression of Parkin,it was found that the mitochondrial autophagy was inhibited,and the antioxidant effect of RES on cells was inhibited,indicating that Parkin-mediated mitophagy plays an important role in the antioxidant effect of RES.In summary,RES increases the activity of antioxidant enzymes in pig intestines by activating Parkin-mediated mitophagy,thereby improving the antioxidant capacity of pig intestines.Gut-healthy feed additives provide a rationale,at the same time,it also provides new ideas and directions for the research on nutritional regulation of alleviating oxidative stress in pig intestines. |
