Dendrocalamus latiflorus is an important dual-purpose bamboo in southern my country.Dendrocalamus latiflorus forest has high ecological and economic benefits.With the development of the Liuzhou river snails rice noodle,the demand for Dendrocalamus latiflorus shoots as an important raw material for sour bamboo shoots has also increased,and the Dendrocalamus latiflorus shoots are facing a shortage of supply.Therefore,in this study,the main branches of Dendrocalamus latiflorus were used as test material.The optimal combination was selected by studying the effects of plant growth regulator type,concentration and soaking time,branch age and cutting time on the rooting of Dendrocalamus latiflorus cuttings.The changes in endogenous hormones,oxidase activity and nutrients during the rooting process were also measured,and the root system was observed anatomically at different periods of time.A preliminary understanding of rooting and physiological substrates for Dendrocalamus latiflorus cuttings to provide a theoretical basis for large-scale factory nurseries.The main findings are as follows.(1)A multi-factor completely randomised test was used to analyse the effects of growth regulator type,concentration and treatment time on Dendrocalamus latiflorus cuttings,and the results showed that : the type of growth regulator had the greatest effect on the rooting of Dendrocalamus latiflorus cuttings,followed by the concentration of the growth regulator and,to a lesser extent,the soaking time.The combinations of IBA(200 mg/L+2 h),NAA(200 mg/L+4 h),IBA+NAA(400 mg/L+2 h),ABT#1(100 mg/L+4 h)and GGR#6(200 mg/L+4 h)treatments were more effective for Dendrocalamus latiflorus cuttings,with rooting rates of 63.33%,68.67%,85.33%,73.33%,and80.00%.A comprehensive analysis of the affiliation function showed that 400mg/L IBA + NAA(1:1)soaked for 2 h was the best treatment for the rooting of Dendrocalamus latiflorus cuttings.(2)There was a significant age effect(P<0.05)for different branch ages on the rooting of Dendrocalamus latiflorus cuttings.The 1 year old branches had the highest rooting rate(49.33%)and the best rooting results,followed by the 2year old branches(43.33%).There was a highly significant effect(P<0.01)of different cutting times on the rooting and survival rates of Dendrocalamus latiflorus cuttings,with the highest rooting and survival rates of 85.33% and81.67% respectively for cuttings taken in mid-March.(3)Observations on the morphological changes in rooting during cuttings of Dendrocalamus latiflorus,The Dendrocalamus latiflorus cuttings go through3 stages of rooting: 0-20 d for root induction,21-60 d for root elongation and after 60 d for root maturation.Dendrocalamus latiflorus cuttings belong to the latent type of adventitious root primordial rooting.(4)The dynamics of endogenous hormones in the spike during the rooting process of Dendrocalamus latiflorus cuttings showed that:The plant growth regulator treatment increased the content of IAA,CTK and GA3,promoting cell division and root elongation;at the same time,it reduced the ABA content and reduced the inhibitory effect of ABA on root growth.The IAA/ABA ratio was always higher in the test group than in the control group,the higher the ratio,the more favourable the rooting.The dynamics of oxidase activity showed that: SOD activity gradually decreased after the root induction period and then increased after 80 d when the shoots sprouted;POD activity increased after the induction period and then decreased after 40 d and stabilised after the root maturation period;IAAO activity increased before 40 d and then slowly decreased after 40 d;PPO activity showed an increasing trend before the root maturation period and then gradually decreased after 60 d.The dynamics of the nutrient content showed that both the soluble sugar content and soluble protein content gradually decreased during the cuttings until60 d and increased after the maturation period.The trend of nutrient decline was greater in the treatment group,with greater depletion than in the control group.
