As a medicine-food homology plant,Codonopsis pilosula is a tonic traditional Chinese medicine.With the increase of demand,the shortage of excellent varieties in C.pilosula planting results in the problem of uneven quality.The study of genuine medicinal material germplasm resources is an important way to improve the quality of Chinese medicinal materials.C.pilosula is an authentic medicinal material in Shanxi,which has the typical "excellent shape" characteristics of "lion head,phoenix tail and chrysanthemum heart".C.pilosula polysaccharide is one of the index components for quality evaluation of C.pilosula,and the content of C.pilosula polysaccharide in Lu Dangshen is significantly higher than that in other producing areas,which is an important embodiment of "high quality" for Lu Dangshen.Therefore,it is of great significance to analyze the mechanism of genuine character formation of C.pilosula at the molecular level for molecular assisted breeding of C.pilosula.In this paper,the correlation between the trait characteristics of Lu Dangshen and Bai Tiaodang introduced by Lu Dangshen and the composition characteristics of C.pilosula polysaccharide was analyzed,and whether the "high quality" and "excellent shape" of C.pilosula were related.On this basis,the transcriptomics database of Lu Party participating in Baitiao Party from different producing areas was constructed,and the related genes related to the formation of "high quality" and "excellent shape" characteristics of Lu Dangshen with different producing areas and Bai Tiaodang were excavated by q RT-PCR technology,which lays a theoretical foundation for analyzing the molecular mechanism of genuine character formation of Lu DangshenThe main results are as follows: 1.By analyzing the traits of Lu Dangshen and Bai Tiaodang from different producing areas,it was found that there were differences in length,diameter under rhizome,adventitious bud number of rhizome head,aboveground branch number and color of Bai Tiaodang participated by.Lu Dangshen Combined with HPGPC analysis of C.pilosula polysaccharide,it was found that the fructan content of Lu Dangshen was significantly higher than that of Bai Tiaodang,and the peak area of fructan at 33 min in C.pilosula HPGPC map was significantly positively correlated with the proportion of annular transverse stripes of Lu Dangshen and the number of adventitious buds in rhizome head(p<0.05);the peak area of fructan at 33 min was negatively correlated with skin L*(D65),skin E*(D65)and wood L*(D65)(p<0.05);the peak area of fructan at 33 min was positively correlated with A*(D65)and B*(D65)(p<0.01),showing that there is a correlation between excellent shape and high quality of Lu Dangshen.2.In order to further analyze the connotation of the correlation between excellent shape and high quality of Lu Dangshen,the transcriptome databases of C.pilosula from six different producing areas were established,and a total of 64.59 G Clean Data were obtained,and the effective data amount of each sample ranged from 9.79 g to11.15 g.A total of 1755 differential genes were screened,of which 922 were up-regulated and 833 were down-regulated in Lu Dangshen.GO enrichment analysis showed that among 818 differentially expressed genes,biological pathway items were the most,followed by cytological components,and molecular function annotated differential genes were the least.KEGG enrichment analysis showed that 202 differentially expressed genes were mainly involved in starch and sucrose metabolism,phenylpropanoid biosynthesis,zeatin biosynthesis,etc.2 pathways,namely starch and sucrose metabolism and zeatin biosynthesis,were related to fructan synthesis and adventitious bud differentiation of reed head,were selected as research targets,and nine candidate genes were screened.3.The expression patterns of candidate genes were analyzed by q RT-PCR,and the Cp GBE gene and Cpβ-glucosidases3 gene which were negatively correlated with the fructan content of C.pilosula were further screened,and the Cp UGT73AH2 gene which was positively correlated with the fructan content of C.pilosula was positively correlated with the fructan content of C.pilosula.Combined with literature research,it is speculated that the Cpβ-glucosidases3 gene was involved in the activation of auxin,while the gene Cp UGT73AH2 was involved in the synthesis of tangshenosideⅠ,atractyloside Ⅲ and zeatin,which lays a theoretical foundation for further verification of gene function and the authentic formation mechanism of C.pilosula.
