Wheat stripe rust caused by obligate biotrophic fungus(Puccinia striiformis f.sp.triici,Pst)is a severe threat to the food security in our country.Excavating disease-re sistant genes and breeding disease-resistant varieties is an effective method of control ing stripe rust.Melatonin and its metabolites play a role in regulating disease resistan ce in plants.Because they can scavenge a lot of H2O2 as antioxidants.In this study,the melatonin synthesis-related genes in wheat were identified,and their expression chara cteristics and functional analysis in the interaction between wheat and stripe rust were explored.The main results are as follows:1.The methyltransferase(MTSs)family of melatonin synthesis-related genes was obtained through systematic identification.The transcriptome data showed that the MTSs gene was differentially expressed at different time points of stripe rust infected.Phylogenetic analysis showed that the genetic relationship between Ta MTSs and Hor MTSs is nearer.2.Sequence analysis clarifies the characteristics of genes.TaMTS1 belongs to the leucine carboxyl methyltransferase family.It contains a LCM functional domain.The promoter region contains cis-acting elements such as ABRE,CGTCA-motif,ERE that regulate plant stress.TaMTS1 was found to be distributed in cytoplasm and nucleus by subcellular localization of tobacco.3.The expression pattern of TaMTS1 in the interaction between wheat and stripe rust was clarified.TaMTS1 was up-regulated at 12 h and 24 h after CYR23 infection.The expression of TaMTS1 was also up-regulated by melatonin,the expression of the disease related protein genes Ta PR1,Ta PR2,and Ta PR5 were up-regulated after melatonin treatment.4.Revealed that TaMTS1 plays a role in disease resistance in the interaction between wheat and stripe rust.There was no significant difference between the treatm ent and control groups after inoculation with CYR31 on the silent plants.After inoculation with CYR23 on the silent plants,the spores produced on the leaves of the silenced plants compared with the control group.Hyphal development accelerated and expression of defense-related genes decreased on the silenced plants by histological observations and accumulation of reactive oxygen species(ROS)increased,the expression of defence-related genes Ta PR1、Ta PR2、Ta PR5 decreased on the silenced plants,which indicated that TaMTS1 knocked down weakened the resistance.
