Dastarcus helophoroides belongs to Coleoptera family.As an important natural enemy of trunk pests,D.helophoroides has been widely used in the control of a variety of pests.After the release of D.helophoroides,they need to search for hosts and may be food shortage during this period.D.helophoroides adults showed a strong hunger tolerance under artificial breeding condition.Trehalose is an important energy storage substance in insect hemolymph,which can regulate the glucose content and plays an important role in maintaining insect activities.In this paper,we explored the starvation tolerance period of D.helophoroides,and screened differentially related expressed trehalose genes under different starvation treatments by transcriptome sequencing.Combined with RNA interference technology,the function of DhTPS gene was verified,which laid a foundation for the optimization of artificial breeding,the establishment of reasonable release strategy and hunger tolerance mechanism in D.helophoroides.The main results are as follows:1.Under the condition of artificial feeding,the mortality reached 45.83% after two weeks of starvation treatment,and the mortality and weight changed significantly.After four weeks of starvation,the mortality reached 97.50%,D.helophoroides showing a strong hunger tolerance.2.61 related trehalose differential genes were obtained by transcriptome data under different starvation treatments.Using RT-QPCR,10 genes were selected to verify the reliability of transcriptome data.3.In the transcriptome data of two starvation and two normal feeding weeks,20 differentially expressed trehalose related genes were screened.In the whole period of hunger(0 week,1 week,2 weeks,3 weeks,4 weeks),DhTPS expression in a certain range;The expression levels of DhTRE-1 and DhTRE-2 decreased;The expression levels of DhTret-3,DhTret-4,DhTret-7,DhTret-14 and DhTret-15 were significantly decreased;The expression levels of DhTret-1 and DhTret-9 were significantly increased.4.when DhTPS interfered for 24 h,gene expression levels of DhTret-5,DhTret-7,DhTret-9,DhTret-10,DhTret-11,DhTret-13 and DhTRE-3 were significantly increased,gene expression levels of DhTret-1,DhTret-2 and DhTret-16 were significantly decreased.D.helophoroides can survive four weeks under starvation stress.And in the process,Adults may respond to starvation by regulating the expression changes of related trehalose genes DhTPS,DhTret-1,DhTret-3,DhTret-4,DhTret-7,DhTret-9,DhTret-14,DhTret-15,DhTRE-1 and DhTRE-2.At the same time,the gene DhTPS has a regulatory relationship with genes DhTret-1,DhTret-2,DhTret-5,DhTret-7,DhTret-9,DhTret-10,DhTret-11 and DhTret-13. |
