| In order to select and breed new stock varieties with good botany trait and good resistance to root knot nematodes(Meloidogyne incognita),On the basis of the identification of cold resistance,drought resistance,salt resistance,alkaline resistance and disease resistance of the progenies of the cross combination of Chinese wild grape and American wild grape(or stock variety)stock in the early stage of the research group,In this study,36 superior plants from 6 combinations were studied:(1)Identification of flowering date,flower type,growth potential,stem yield and survival rate of cuttage propagation of superior plants of 4 rootstock hybrid combinations was carried out.(2)The root-knot nematodes in diseased melon roots were isolated and identified as root-knot nematodes(Meloidogyne incognita)by morphology and molecular biology.The potted plants of the test materials were inoculated with melon root knots containing root knot nematodes(Meloidogyne incognita),and the potted plants of rootstock variety ‘5BB’ and cultivated cultivar ‘Summer Black’ were used as control,after 80 days,their resistance performance was evaluated according to disease index,and the results were as follows.1.The first flowering of the progenies of the four crosses was in early May in Yangling,Shaanxi province,(V.riparia×V.amurensis Zuoshan 75097)combination offspring female flowers: male flowers =3:1,(V.amurensis Taishan-11×V.riparia Mcadams)combination offspring Female flowers: male flowers=3:5,(V.amurensis Taishan-11×V.rupestris)combination offspring Female flowers: male flowers: bisexual flowers =4:8:1,and(‘5BB’×V.amurensis Heilongjiang seedling)offspring Female flowers: Male flowers: bisexual flowers=6:7:2.2.In the 4 combinations,except the hard branch cutting survival rate of the descendant lines of(V.amurensis Taishan-11×V.rupestris)was low,the survival rate of hard cuttings of the descendant lines of(V.riparia×V.amurensis Zuoshan 75097),(V.amurensis Taishan-11×V.riparia Mcadams),and(‘5BB’×V.amurensis Heilongjiang seedling)was more than 60%,and most of them were above 70%.3.The growth potential of 26 lines of 4 rootstock hybrid combinations was evaluated by taking the height,stem diameter and number of leaves of cuttage seedlings as indexes,and combining with the growth potential of mature trees of 8 year old hybrid superior plants,The results showed that there were 11 strains with strong growth potential : HS-2,HS-9,HS-10,HS-12,HS-13,TH-1,TH-5,TH-18,BBS-13,BBS-14 and BBS-17.There were 10 lines with moderate growth potential : HS-1,HS-3,TH-7,TS-3,BBS-1,BBS-2,BBS-3,BBS-9,BBS-10 and BBS-16.The other five strains grew weakly.4.The resistance of 36 materials from 6 grape rootstock combinations to root-knot nematodes was tested.The results were as follows: Immune 14 strains: ‘5BB’(CK1),HS-2,BBS-1,BBS-2,BBS-4,BBS-5,BBS-7,BBS-9,BBS-10,BBS-11,BBS-12,BBS-15,BBS-16,BBS-17 and 19-4-3.High resistance 2 strains : HS-5 and TH-1.Resistance 5strains : HS-1,TH-18,TS-21,BBS-13 and BBS-14.Middle resistance 3 strains : HS-4,HS-9and16-2-5.Susceptibility 4 strains : HS-3,HS-10,13-5-2 and 16-2-7.High susceptibility 8strains: ‘Summer Black’(CK2),HS-12,HS-13,TH-5,TH-7,TH-11,TS-3,TS-4,and 17-2-1.Among them,the resistance of the progeny of combination(‘5BB’×V.amurensis‘Heilongjiang seedling’)was more inclined to immune or high resistance,accounting for86% and 14% respectively.5.The results showed that HS-2,BBS-17 and TH-1 had good botanical character and showed immune or high resistance to root-knot nematode,followed by HS-1,TH-18,BBS-13 and BBS-14 had good botanical characters and were resistant to root-knot nematodes. |
