Objective: To explore the mechanism of continuous cropping obstacles and mitigation methods of Pinellia ternate(Thunb.)Breit.in order to solve the problem of continuous cropping obstacles that existed in P.ternate production,and to guide the industrialized cultivation of P.ternate.Methods: Dynamic monitoring different joints and different modified measures(2% bio-charcoal,bio-organic fertilizer,microbial bacteria and grass ash)lower P.ternate growth index and soil physicochemical indicators,using Pacbio Sequel sequencing platform for P.ternate steady soil microorganisms Sequencing;and the content of total alkaloids and soluble proteins were determined by ultraviolet spectrophotometry,HPLC method to determine the content of gobylide and adenosine;and primarily for P.ternate,Determination of extract content of P.ternate by cold soaking and using Q-Orbitrap High resolution liquid quality Usage,measuring the P.ternate stempolic acid chemical substance.Results:(1)Compared with the first crop of soil,the bulk density and water content of the second crop and the third crop showed a continuous increase trend,while the soil organic matter showed a continuous decline trend;as the number of continuous cropping stubble increased,the total nitrogen,Available nitrogen and total phosphorus showed a continuous downward trend,available phosphorus and total potassium showed a trend of first increasing and then decreasing,and available potassium continued to increase.(2)With the increase of continuous cropping stubble,the diversity of bacteria and fungi in the rhizosphere soil of P.ternate decreased.The abundance of beneficial fungi such as the Glomeromycota flora decreases,and the abundance of potential pathogenic bacteria increases.If Pectobacterium aroidearum,potential pathogenic fungi such as Ascomycota flora,Fusarium nematophilum and Fusarium nematophilum The abundance of Thanatephorus cucumeris has increased.(3)With the increase in the number of continuous cropping crops,the output of P.ternate gradually declines.Compared with the first croping(CK),the first continuous cropping(T1),second continuous cropping(T2)and third continuous cropping(T3)decreased by 11.97%,57.12% and 68.68% respectively;compared with CK,the extracts of P.ternate in second continuous cropping(T2)and third continuous cropping(T3)The content increased by 4.57% and 14.93%,respectively,while the first continuous cropping(T1)treatment decreased by 13.78%;compared with the first croping(CK),the alkaloid content of the first continuous cropping(T1)and third continuous cropping(T3)increased by 24.28% and 11.39%,and second continuous cropping(T2)decreased by 11.39%;compared with the first croping(CK)In comparison,the soluble protein content of the first continuous cropping(T1),second continuous cropping(T2)and third continuous cropping(T3)increased by 18.11%,38.64% and 34.82%,respectively.(4)Microbacterial agent,biological organic fertilizer,and grass ash can promote the growth of P.ternate,the development of the tubers and the growth of the root system;the impact on the products of P.ternate,no significant difference,single production analysis,the effect is best in the grass ash Biological organic fat.(5)P.ternate tuberculous,P.ternate,ethyl acetate extract,exhibited high concentration and low concentration promoted germination,medium concentration inhibited germination,and then extracted with ethyl acetate after extracting water purifiers to P.ternate The germination plays a high concentration inhibitory and low concentration promotes germination.The P.ternate stems have been found through different extraction methods,and extracts reflect different degrees of chemical inhibitory effects,and the LC-MS detection of different treatments has found that all extracts contain Nitbo gold ispropyl esters and Kaboba.Conclusion With the increase of consecutive stubble times of P.ternate,the physical and chemical properties of the soil change,the abundance of beneficial microorganisms in the rhizosphere soil decreases and the abundance of harmful microorganisms increases,the diversity of rhizosphere soil microbial communities decreases,and the p H fluctuation of the rhizosphere soil changes.,Is an important reason for the formation of P.ternate continuous cropping obstacles.Bio-organic fertilizer and plant ash can alleviate the field effect of continuous cropping obstacles of P.ternate. |
