In this experiment,the coarse powder of each part of Sophora flavescens was used as the test material,and the crude extract was extracted from each part by ultrasonic extraction method;According to the inhibition effect of the n-butanol extract of Sophora flavescens root,the content of matrine in the root of Sophora flavescens was quantitatively analyzed,and the optimal extraction conditions of matrine were fitted by response surface analysis method according to the content of matrine;Six kinds of pesticides were screened for indoor bacteriostasis,and the two pesticides with the best inhibitory effect were selected for compound research with 1.3%matrine water preparation,and field and indoor control effect tests were carried out;The antibacterial mechanism of the test liquid was preliminarily studied by permeability,three enzyme activities and soluble protein content,and the results showed that:1.The inhibitory effect of extracts from various parts of Sophora flavescens on two ginseng pathogens is from high to bottom:Sophora Radix>Sophora Radix Root Bark>Sophora Radix Seeds>Sophorae Radix Sophorae Radix>Sophorae Radix et Rhizoma Leaf The sequence is n-butanol,chloroform,ethyl acetate,acetone,methanol,and ethanol.2.The optimal extraction conditions of matrine are n-butanol as the extractant,the ratio of the solid-liquid ratio of Sophora flavescens coarse powder to n-butanol is1:10.02,the extraction is repeated 3 times,the extraction temperature is 48.90℃,and the total extraction is 1.54 h,the theoretical value of matrine extraction was 11.2583mg·g-1.3.Six pesticides were selected to carry out antibacterial tests on the two types of bacteria.The two types of ginseng black spot bacteria with the best inhibitory effect were polyantimycin B and difenoconazole.The best two are propiconazole and chlorothalonil.4.In the antibacterial test of ginseng black spot disease,when the compound ratio of 1.3%matrine water and difenoconazole is 5:5,the toxicity ratio is 1.19,and 1.3%matrine water and polyvinyl When the compounding ratio of prime B is 7:3,the virulence ratio is 1.15,and the additive effect is obvious;in the antibacterial test of ginseng botrytis cinerea,when the compounding ratio of 1.3%matrine water and propiconazole is 9:1,the toxicity ratio is 1.17,when the compound ratio of 1.3%matrine aqueous solution and chlorothalonil is 5:5,the toxicity ratio is 1.16,which is an additive effect.After mixing according to the above four proportions,the co-toxicity coefficients were 225.66,299.24,305.93 and 259.23,all of which were greater than100,which was an obvious synergistic effect.5.The control effect test showed that after the indoor compound treatment of leaves,the lesion area was the smallest,and the control effect on the two diseases of ginseng after the field compound treatment was significantly better than that after the single-agent treatment,which verified the indoor bacteriostatic results.After the combination of matrine water and pesticide,the effect on two diseases of ginseng has been significantly improved6.Preliminary research on antibacterial mechanism shows that after the treatment with the agent,the mycelium has obvious extravasation of the cytosol,the electrical conductivity gradually increases,the cells are effectively destroyed,and the growth of the mycelium is effectively inhibited.Enzyme(SOD),peroxidase(POD),catalase(CAT)three important protective enzymes were measured,and it was found that the activities of the three enzymes could be effectively reduced,and the synthesis of soluble protein content was inhibited at 8 h effect.
