China is a big potato production country in the world.As the four major food crops,potato is playing an increasingly important role in ensuring national food security.Carotenoids have important health care functions and are closely related to human health.Screening and breeding new potato germplasm with high Carotenoid will help to improve human health and quality of life.However,the carotenoid content of potato will change with storage conditions.Therefore,it is of great significance to analyze the change trend of carotenoid content under different storage conditions,and then to screen high-carotenoid potato germplasm.For this reason,this study selected 52 colorful potatoes as the test materials,different storage conditions at normal temperature(light avoidance room,contrast)and low temperature(cellar,5~10 ℃),by measuring the total carotenoid content in tubers during 0~60 d storage,as well as anthocyanins,starch,reducing sugars,and vitamins C and other quality index content and total antioxidant capacity,further analyze the change trend of potato tuber carotenoid content and other related indicators,and through carotenoid-related gene expression analysis,screen out potato varieties with high carotenoid content,main conclusions as follows:further analysis of the change trend of potato tuber carotenoid content and other related indicators,and,(1)The colorimetric method was used to determine the carotenoid content of potato under different storage conditions.The results showed that: 1)The carotenoid content in tubers of different varieties of potato is significantly different;2)Under different temperature storage conditions for 60 d,the change trend of carotenoid content in tubers of most potato varieties is increasing,and the content of a small part decreases;3)When stored for 60 d,the content under low temperature crypt treatment is higher than that after normal temperature room treatment,but there is no significant difference;4)Under 60 d storage conditions,the variety and temperature have significant differences in the content of tuber carotenoids(P<0.05).(2)Under different storage conditions of 60 d,the change trend of anthocyanin content of potato varieties has been decreasing,and the content after low temperature treatment is higher than that after normal temperature treatment under different temperature treatments during the same storage period.Storage has a better effect on delaying the degradation of anthocyanins of potato tubers than storage at room temperature.The change trend of the total antioxidant capacity of potato varieties under different storage conditions for 60 days is mainly increased.During the same storage period,the total antioxidant capacity of tuber after low temperature treatment is not significantly different from normal temperature treatment;at 60 d,potato total antioxidant capacity is significantly associated with total carotenoids,and the late storage capacity is associated with carotenoid content with the extension of storage time.(3)The change trend of quality index of 52 potato varieties stored under different conditions for 0~60d: 1)The change trend of tuber starch content is that the content decrease.When stored for 60 d,the content under low temperature treatment is higher than that under normal temperature treatment.2)The changing trend of reducing sugar content in potato tubers is increasing,which is opposite to the changing trend of starch content and is negatively correlated.And the varieties with low reducing sugar content in potato tubers under different treatments are mainly processed potatoes.3)The change trend of potato tuber protein content is increasing.When stored for 60 days,the content under low temperature treatment is higher than that under normal temperature treatment,the content change is small,the difference is not significant,and it is a relatively stable index.At the same time,it was found that the protein content of purple skin and purple flesh type potatoes was higher than that of other types of potatoes in the early storage period.4)Vitamin C is an extremely unstable indicator.The change trend of the vitamin C content of 44 potato tubers has been decreasing,and the decrease is particularly large;after 60 days of storage,the vitamin C content of most varieties is the content after storage at room temperature Higher than low temperature storage,fresh potatoes should be eaten as soon as possible in terms of the vitamin C content of potatoes.(4)Using potato leaves at the seedling stage to analyze the expression of carotenoid-related genes,the results showed that the relative expression levels of PSY,PDS and LYC-e in potatoes with white flesh were too low,and relative expression levels in potatoes with yellow flesh High is positively correlated with the total carotenoid content of tubers,while the expression level of LYC-b gene is opposite to the total carotenoid content and is negatively correlated.(5)Comprehensive changes in the content of various quality indicators,60 d short-term storage can choose room temperature storage,long-term storage is suitable for low-temperature storage;high-carotenoid potato varieties Huasong 7 and Xisen 6 were selected,which can be used as Parental material for breeding new potato varieties. |
