| Skeletal muscle is the largest and most widely distributed organ in the body.People’s daily life activities cannot do without the support and cooperation of skeletal muscle.After skeletal muscle injury,the body initiates a repair program.Since the self-healing process is slow and the results are usually not ideal,people are committed to trying different methods to promote skeletal muscle repair and regeneration.As a traditional Chinese medicine for promoting blood circulation and removing blood stasis,Danshen is often used in combination with other Chinese medicines to promote muscle fiber repair.However,there are no relatively comprehensive reports on the effect of Danshensu on skeletal muscle regeneration.In this experiment,a mouse injury model was established and Danshensu was used to treat.Histological observations and molecular analysis of the therapeutic effects were carried out from different time periods.It is hoped that the mechanism of Danshensu’s effect on skeletal muscle regeneration and fibrosis can be discovered,thereby promoting animal husbandry.The growth and development of livestock species and the quality of meat products will improve the economic benefits of breeding animals.The results of the study are as follows:(1)Use Evans blue staining and HE staining to determine the best dose and the best observation period for the establishment of the damage model.Fluorescence quantitative PCR technology was used to detect the gene expression changes of muscle growth-related factors and myostatin in mice after injury,and it was determined that the mice were successfully injured.(2)Through Evans blue staining and HE staining,the positive effects of Danshensu treatment on the regeneration of damaged muscles were observed.Through fluorescence quantitative PCR technology and Western blot technology,the detection of muscle regeneration-related factors,macrophages and inflammatory factors,new muscle fibers,and fat marker gene expression showed that Danshensu treatment can upregulate the positive regulation of muscle growth.Factors Myo D and Myo G inhibit the expression of myostatin MSTN.It can also promote the activation and proliferation of muscle satellite cells,intervene in the inflammatory response to provide a good regeneration environment for muscle fibers,enhance fat production in damaged muscles,and further accelerate the regeneration and repair process of skeletal muscle.(3)Through Masson staining,the inhibitory effect of Danshensu treatment on fibrosis after regeneration of damaged muscles was observed.And using fluorescent quantitative PCR technology and Western blot technology to detect the expression of type I collagen fibers and myofibroblast marker factorα-SMA,it was found that the expression of fibrosis-related genes in the treatment group was lower than that in the injury group.It shows that Danshensu treatment can effectively inhibit the formation of collagen fibers in the process of muscle repair,effectively slow down the formation of scar tissue,and further improve the quality of skeletal muscle regeneration.The results of this study show that Danshensu,as an effective monomer component in Danshen,induces skeletal muscle regeneration from many aspects,and while accelerating the repair speed,it also improves the quality of skeletal muscle repair and strengthens the skeletal muscle during the healing process.Competitiveness of repair and regeneration for fibrotic scars. |
