Leptocybe invasa Fisher&La Salle is a kind of exotic invasive gall insect that seriously harms the branches and leaves of Eucalyptus,causing great economic loss to the development of Eucalyptus industry.Traditional microbial isolation and culture combined with 16 S r DNA gene sequencing and Illumina Hi Seq high-throughput sequencing technology were used for the study on the effects of winter and summer and different host plants on the community composition and diversity of bacteria in female adults of L.invasa,the similarity of symbiotic bacteria in DH201-2(E.grandis×E.tereticornis)-L.invasa-Q.mendeli was studied.It can lay a foundation for exploring the physiological function,sources and transmission pathways of symbiotic bacteria in L.invasa.The research results are as follows:(1)Seventeen different strains of bacteria were isolated from summer population,belonging to 15 genera,among which Firmicutes and Proteobacteria were the dominant bacterium,and Staphylococcus,Pantoea were the dominant genus.11 strains of bacteria were isolated from winter population,belonging to8 genera,among which Firmicutes was the dominant bacterium,and Bacillus was the dominant genus.The richness and diversity of culturable bacteria in summer population were higher than winter.The species composition and diversity of culturable bacteria in female adults of L.invasa were different in winter and summer.(2)Actinobacteria and Proteobacteria were the dominant bacterium of culturable bacteria in L.invasa from different host,but Staphylococcus,Pantoea were the dominant genus in female adults of L.invasa feeding on DH201-2(E.grandis× E.tereticornis),while Curtobacterium was dominant genus in the population feeding on E.exserta.The culturable bacteria richness and diversity of the population feeding on DH201-2(E.grandis×E.tereticornis)were higher than that feeding on E.exserta.Food changed the diversity of culturable bacteria in L.invasa.(3)The species richness of bacteria in L.invasa feeding on DH201-2(E.grandis×E.tereticornis)was higher,but the diversity was lower.At the phylum level,the dominant bacterium was Proteobacteria in the both populations.However,there were a few new taxa that were not classified in the population feeding on DH201-2(E.grandis× E.tereticornis).At the genus level,the microflora of L.invasa feeding on DH201-2(E.grandis×E.tereticornis)were mainly composed of Rickettsia(93.11 %).while the microflora of L.invasa feeding on E.exserta is mainly composed of Rickettsia(73.61%),Pantoea(10.84%)and Rhizobium(8.01%).Host plant was one of the factors affecting the molecular diversity of bacterial community in the female adult of L.invasa.(4)Bacillus,Staphylococcus and Panbacteria were found in the larvae and the L.invasa.Bacillus was found in all 7 tissues,and Curtobacterium was found in DH201-2(E.grandis×E.tereticornis),gall tissues,L.invasa and Q.mendeli.Panbacteria was found in larvae,L.invasa and Q.mendeli;L.invasa and Q.mendeli contained Pseudomonas,Stenotropho monas and so on.The culturable bacteria in Eucalyptus-L.invasa-Q.mendeli had similarities.(5)The richness and diversity of bacteria in DH201-2(E.grandis×E.tereticornis)and gall tissues were higher than those in other tissues,but the composition of bacteria was similar.Proteobacteria and Firmicutes were the dominant bacterium and Methylobacterium was the dominant genus in the bacterium community of DH201-2(E.grandis×E.tereticornis).Proteobacteria and Firmicutes were the dominant bacterium and Streptococcus was the dominant genus in the bacterium community of gall tissues.Proteobacteria was the dominant bacterium and Streptococcus was the dominant genus in the bacteria of the larvae and adults of L.invasa in nanning area and L.invasa and Q.mendeli in sichuan area.There were 11 identical phyla and 96 identical genera in the bacteria of the larvae and adults of L.invasa,and the proportion of similar species is higher.There were 9 identical phyla and 78 identical genera in the bacteria of DH201-2(E.grandis×E.tereticornis),gall tissues,the larvae and adults of L.invasa,which had some similarities with each other.There were 10 identical phyla and 98 identical genera in the bacteria of L.invasa in nanning and sichuan area,and 11 phyla and 121 genera were shared by L.invasa and Q.mendeli in sichuan area.There is a close relationship between eucalyptus,L.invasa,and Q.mendeli.The winter and summer seasons and host plants are important factors affecting the composition and diversity of bacterial communities in L.invasa.The bacteria in eucalyptus,L.invasa and Q.mendeli were similar,indicating that there is a close relationship among them.
