Tirathaba rufivena(Walker)(Lepidoptera: Pyralidae)is an insect that causes serious damage in palm plants.Its transcriptome and protein resources are still in the blank stage,which confines our research on its growth,development and prevention at the molecular level.In this study,we sequenced the full-length transcriptome of the insect using PacBio Iso-Seq in combined with Illumina RNA-Seq and examined the differentially expressed genes(DEGs)at different developmental stages(egg,larva,female pupa,male pupa,female adult and male adult).The main results are as follows:1.Based on PacBio Sequel II and Illumina Nova Seq 6000 sequencing platforms,approximately 31.26 Gb subreads were obtained by full-length transcriptome sequencing and 746 millions 150 bp high-quality clean reads were generated by transcriptome sequencing at different developmental stages.Including 236,575 circular consensus sequence(CCS)and 180,244 full-length non-chimeric(FLNC)subreads.After clustering and correction,39,228 consensus isoforms,including 39,199 polished high-quality ones,were identified.A total of 38,390 transcript isoforms were obtained by removing redundant subreads.The average length of transcript isoforms was 2,304 bp with N50 of 3,310 bp.2.The non-redundant transcript isoforms were BLAST analyzed against four functional databases,form which 31,821 isoforms were annotated.Our analysis predicted a total of 31,948 coding sequences(CDS),3,450 alternative splicing(AS)events,527 transcription factors(TFs),8,511 simple sequence repeats(SSRs)and 5,077 Long non-coding RNAs(lnc RNAs).In addition,we observed 26,209 DEGs at different developmental stages of T.rufivena.These annotated isoforms provide the molecular data basis for the development of gene regulation,phylogenetic evolution,growth and development,physiological metabolism,and large-scale molecular markers of T.rufivena.3.Based on annotated transcripts,415 detoxification-related and 29 sex determination-related candidate transcripts were predicted and their expression patterns at different developmental stages were investigated.Our study provides a new and comprehensive information on the transcriptomics of T.rufivena,which will be useful for further study on its gene expression,insect development,and pest control.
