| In vitro maturation and in vitro fertilization of oocytes are the main technical means of in vitro embryo production and reproductive biology research.They play a very important role in the current production and scientific fields,but the in vitro culture system of oocytes still needs to be improved,affecting oocyte the external factors of the quality of cell maturity,especially the problem of oxidative damage,still need to be solved.Epigallocatechin gallate(EGCG)has strong antioxidant properties,but it still lacks systematic research on the effect of in vitro maturation and in vitro fertilization of porcine oocytes.Therefore,this study took pig oocytes as the research object,by adding different concentrations of EGCG treatment,to observe its effect on the in vitro maturation and in vitro fertilization of pig oocytes,combined with the expression of related genes and antioxidant indicators to explore EGCG The effects on the in vitro maturation and in vitro fertilization of porcine oocytes and their possible mechanism of action provide a theoretical basis for further improving the in vitro maturation culture system of porcine oocytes.First,EGCG(0 μM,5 μM,10 μM,20 μM)was added to the in vitro maturation solution of porcine oocytes to culture porcine oocytes.The results showed that the cumulus expansion index and maturity rate of the 10 μM EGCG treatment group were significantly higher in the control group(P<0.05),the 10 μM and 20 μM EGCG treatment groups significantly reduced the cellular ROS level compared to the control group(P<0.05).Second,compared with the control group,the total antioxidant capacity of oocytes in the 10μM EGCG-treated group was significantly increased(P<0.05),glutathione content was significantly increased(P<0.05),and propylene glycol content was significantly reduced(P<0.05).The expression of SOD,CAT and GPx mRNA in the antioxidant genes of oocytes in the 10 μM EGCG treatment group was significantly higher than that in the control group(P<0.05).Compared with the control group,the expression of the apoptosis gene Casepase-3 in the 10μM EGCG treatment group was reduced,the expression of the gene Bax was significantly reduced(P<0.05),and the expression of the gene Bcl-2 was significantly increased(P<0.05).In addition,the cleavage rate and blastocyst rate of oocytes treated with 10 μM EGCG added to the mature fluid and fertilization fluid were significantly higher than those of the control group(P<0.05).In summary,the results of this experiment show that the addition of 10 μM EGCG in the IVM stage and IVF stage has a synergistic effect,which can effectively improve the maturation and fertilization of oocytes in vitro,and effectively improve the antioxidant capacity and intracellular GSH concentration of oocytes.And reduce the level of lipid oxidation in oocytes,while regulating the expression of antioxidant genes and apoptosis genes during oocyte maturation. |
