In order to compare and analyze the differences in immune response between the two color types Channa argus,they are divided into B-PBS,B-Ah,W-PBS and W-Ah conducted an Aeromonas hydrophila infection experiment.The B-PBS and W-PBS groups were injected with 100μl PBS,and the B-Ah and W-Ah groups were injected with the same volume of 3.6×10~5 CFU/ml A.hydrophila.The mortality rate of each group was counted separately,the changes of plasma biochemical indexes and intestinal tissue morphology,and the expression of immune-related genes were analyzed;and the transcriptome technique was used to analyze the differences in immune responses between the two color types Channa argus.The test results are as follows:(1)After injection of 3.6×105 CFU/m L of Aeromonas hydrophila,there was no death in the B-PBS and W-PBS groups,and the mortality rate in the W-Ah and B-Ah groups showed a trend of increasing first and then decreasing The mortality rate was the largest at24 hours,and the mortality rate in the W-Ah group was higher than that in the B-Ah group.(2)The content of plasma malondialdehyde(MDA)in the B-Ah and W-Ah groups showed a trend of increasing first and then decreasing,while superoxide dismutase(SOD)and total antioxidant capacity(T-AOC)were in contrast.Compared with the W-PBS group,the MDA in the W-Ah group was significantly different at 3 h,12 h,and 24 h(P<0.05),and the SOD content in the W-Ah group was significantly different from 6 h to 96 h(P<0.05).The plasma T-AOC content in the W-Ah group was significantly different from 6 h to 48 h(P<0.05).Compared with the B-PBS group,the plasma MDA and T-AOC levels in the B-Ah group were significantly different at 12 h(P<0.05),and the plasma SOD levels in the B-Ah group were significantly different from 24 h to 96 h(P<0.05).(3)The intestinal villi in the B-PBS and W-PBS groups were intact.The intestinal villi showed rupture and shedding at the 24th hour in the B-Ah and W-Ah groups.At 24 h and 96h,the intestinal villi length and density were smaller in the W-Ah group than in the B-Ah group.(4)The expressions of IL-1βand IL-8 in the B-Ah and W-Ah groups were up-regulated,while the expression of IL-10 was down-regulated.The intestinal change trend is basically the same as the liver change trend.Compared with the W-PBS group,IL-1βand IL-8 in the liver were significantly increased in the W-Ah group from 3 h to 48 h(P<0.05),and IL-10was in the W-Ah group from 6 h to 48 h.The difference was significant(P<0.05).In the W-Ah group,IL-1βwas significantly increased at 3 h(P<0.05),and IL-8 and IL-10 were significantly different at 3 h and 6 h(P<0.05).Compared with the B-PBS group,the expression levels of IL-1β,IL-8 and IL-10 in the liver of the B-Ah group were significantly different at 12 and 24 h(P<0.05),and the IL-1βin the skin of the B-Ah group,IL-8 and IL-10 were significantly different at 3 h(P<0.05).(5)By analyzing the liver transcriptome data of C.argus,it was found that the differential genes of C.argus after infection with A.hydrophila were mainly enriched in the complement and coagulation cascade in the immune system.Therefore,after the infection of A.hydrophila,the oxidative stress,intestinal damage and inflammatory reaction suffered by the White type C.argus are more severe than that of C.argus,and the antioxidant capacity is lower.It is speculated that the difference in immune response between the two color types C.argus is controlled by the complement system in the cellular process.
