| The genetic improvement of abdominal fat percentage in broilers has been the difficulty of breeding in broilers.Because the abdominal fat percentage and abdominal fat weight are difficult accurately observe in vivo,traditional breeding methods makes less progress in these characters.In this study,we selected the key genes that influenced the abdominal fat deposition of slow-type yellow feather broilers by the early screening,and we analyzed the influence between gene mutations and the abdominal fat percentage of slowtype yellow feather broilers.Finally,we identified the SNPs which associated with abdominal fat percentage in order to lay the foundation for molecular marker assisted selection in broilers.In this study,we selected 11 candidate genes which related to the abdominal fat deposition in slow-type yellow feather broilers.56 high-and low-abdominal fat percentage samples of5 breeds which include Guangxi Lingshan Chicken,Bantam Chicken,Ephedra Chicken,Xinghua Chicken,and Recessive White Feather Rock Chicken were selected.And we find SNPs form the candidate gene fragments which have been sequencing after amplification by PCR.Then 100-day-old Guangxi Lingshan chicken population with abdominal fat percentage records have been chosen as the material.Genotyping the candidate SNPs in this population by MALDI-TOF and analysis the correlation between SNPs and abdominal fat percentage.The main results are as follows:(1)There were 56 samples with high or low abdominal fat percentage which were selected from 5 breeds.We found 63 candidate SNP sites in candidate fragments form sequence.(2)Genotyping the candidate SNPs in this population by MALDI-TOF in Lingshan chicken which have been recorded abdominal fat percentage.After excluding those SNP sites that failed to genotyping and the minimum allele frequency was less than 0.05,deviated from the Hardy-Weinberg equilibrium,we have analyzed the population genetic parameters of 46 candidate SNPs.The polymorphism information content of 36 SNPs was between 0.25 to 0.5,which belongs to moderate polymorphic sites.(3)Analyzed the correlation between SNPs and abdominal fat percentage in Guangxi Lingshan chicken by single factor analysis of variance model of SPSS 21.0.The result showed that 12 SNPs were significantly related to abdominal fat percentage(P<0.05).These significantly SNPs are located among 7candidate genes,such as: ACBD5,ACSL6,APOA5,APOB,FABP1,LPIN1,SLC27A2。Among the significant SNPs,there are 2 SNPs located in the 3’UTR of ACBD5 and ACSL6.There are 2 SNPs located in the intron of LPIN1 and 4SNPs are located in the intron of FABP1,the other 2 SNPs are located in the intron of SLC27A2.In the exon 3 of APOA5,the allele in c.459G>A mutation into allele A has changed the code which result in the coding amino acid change from glutathione to lysine.In the exon29 of APOB,the allele in c.246C>T mutation into allele T has changed the code which result in the coding amino acid change from glutathione to histidine. |
