| Glume is an important part of rice floral organs,which affects rice yield and quality.In our previous study,an indica rice variety Minhui 3301 was mutagenized by EMS,and a glume mutant,temporarily named bah(beak abnormal hull),was found in the progenies.The structure of bah showed abnormal,some of the lemma incurvate and looks like beak-shaped,the smaller palea shrinking to lemma or missing body of palea,only 2 pieces of transparent marginal region of palea,or some of the lemma and palea were normal but their grains became shorter andnarrower.The genetic mechanism of underlining beak-shaped abnormal hull mutant is not clear yet,so the phenotypic identification,genetic analysis and gene mapping of bah glume mutant were studied,and the results will lay the foundation for further cloning and functional study of BAH.The main results of this study are shown as follows:1.There were no significant differences in plant height,the length of flag leaf and top second leaf between bah and wild type Minhui 3301 at mature stage.However,for bah mutant,the flag leaf and the top second leaf became wider,panicle length shorter,the number of effective panicles per plant lower,the number of spikelet per panicle less,seed setting rate lower,1000-grain weight smaller,and the pollen sterility rate higher,showing a significant or very closely significant differences compared to the wild type.2.From the stamen primordium middle stage,the palea development of bah was slightly slower than that of the wild type,which affects the process of palea development.Therefore,shrinking of the palea within BAH may be due to the slow development of the palea,resulting in a decrease in the bah palea.3.The glume mutant trait of bah showed recessive and was controlled by one pair of recessive genes.BAH was located on chromosome 9,with about 379kb of the physical interval distance between the SSR markers RM24687 and RM24701.4.Through the analysis of the annotated genes in the mapping interval of bah and the sequencing comparison of TCP family transcription factors REP1and Os09g0521300,it was found that there was no difference in the sequence for the two genes between bah and wild-type Minhui 3301,so the two genes were not the target genes of bah.According to the results of this study,it is speculated that BAH may be a new functional gene that controls floret development in rice. |
