Maize(Zea mays L.),as one of the most important food and forage crops in China,has important economic value.In agricultural production,mechanical harvesting can reduce labor costs,and corn mechanical harvesting has high requirements for corn varieties,especially the mechanical harvesting varieties with the characteristics of fast dehydration rate and good lodging resistance.Abscisic acid(ABA)is an essential hormone for plant growth and development.It is involved in the regulation of various physiological processes in plants,such as embryo development and seed dormancy.It plays an important role in plant development and abiotic stress response.In Arabidopsis,ABA can be sensed by PYLs(ABA receptors),resulting in PP2 C inactivation and Sn RK2 activation,and regulating downstream gene expression through signal transduction pathways.However,the study on ABA signal transduction components in maize is still unclear.In this study,the protein localization and interaction network of ZmPP2 Cs and ZmSnRK2 s in ABA signal transduction pathway were analyzed by bioinformatics analysis,subcellular localization analysis and interaction result analysis,so as to provide theoretical basis for the future study of ABA signal pathway in maize.The main results are as follows:1.Bioinformatics analysis: By comparing the PP2 C family and Sn RK2 family in Arabidopsis,15 ZmPP2 Cs and 14 ZmSnRK2 s with high homology with Arabidopsis were screened out and named as ZmPP2 Cs and ZmSnRK2 s.2.q RT-PCR results showed that ZmPP2C1/2/4/6/7 was constitutively expressed in all tissues of maize,the other ZmPP2 Cs were specifically expressed in embryo and endosperm of maize,and ZmSnRK2 s was constitutively expressed in all tissues of maize.3.The results of ZmPP2 Cs subcellular localization showed that ZmPP2C8,ZmPP2C11,ZmPP2C12 were localized in the nucleus.ZmPP2C3/6/13/16 was localized in the nucleus and cytoplasm.and ZmPP2C1/2/4/7/9/10/14/15 was localized in the cytoplasm.4.The results of ZmSnRK2 S subcellular localization showed that:ZmSnRK2.1/3/4/5/6/9/10/13/14 of the ZmSnRK2 family proteins are localized in the cytoplasm and nucleus.ZmSnRK2.2/7/8/11/12 was localized in the cytoplasm.5.The results of bimolecular fluorescence complementation assay(Bi FC)showed that ZmPP2C3 interacted with all ZmSnRK2 s except ZmSnRK2.2/7,and acted in the nucleus.ZmPP2C6 interacts with ZmSnRK2.5/10 in the nucleus.ZmPP2C8 interacts with ZmSnRK2.2/4/6/8/10 in the nucleus.ZmPP2C12 interacts with ZmSnRK2.9 in the nucleus and with ZmSnRK2.12 in the cytoplasm and nucleus.Except for ZmPP2C3/6/8/12/13,the rest of ZmPP2 Cs interacted with some of ZmSnRK2 s in the cytoplasm,but ZmPP2C13 did not interact with all of ZmSnRK2 s.In summary,we studied the gene cloning,expression,protein subcellular localization and interaction of ZmPP2 Cs and ZmSnRK2 s families.These results provide a theoretical basis for further study on ABA signal transduction regulation in monocotyledonous maize.
