Naematelia aurantialba,also known as yellow fungus,brain ear,is a famous rare edible and medicinal fungus in my country.Naematelia aurantialba are unique in color,fragrance and taste,and are of high nutritional and health-care medicinal value.They are favored by consumers.The product is in short supply and has a huge market potential.The high market price of Naematelia aurantialba and good cultivation efficiency have stimulated the cultivation enthusiasm of mushroom farmers and promoted the development of the industry.The Naematelia aurantialba fungus species used for production are composed of two kinds of hyphae of Naematelia aurantialba and hairy leather fungus,and only when the Naematelia aurantialba hyphae reaches a certain proportion,high yield and high quality of Naematelia aurantialba can be realized.In the production process of Naematelia aurantialba,the low ratio of Naematelia aurantialba hyphae in the strains often leads to reduced production or even no harvest.Due to the technical bottleneck of Naematelia aurantialba,the industry develops slowly.Therefore,it is particularly necessary to carry out research on the identification and preparation of effective strains of Naematelia aurantialba.In this experiment,fresh Naematelia aurantialba fruit bodies are used as the test materials,and the research on the identification technology of effective Naematelia aurantialba bacteria is the starting point.The different parts of the longitudinal section of the fruit bodies are identified by micromorphology and molecular biology,and the different fruit bodies are separated Identify the strains obtained from the parts,determine the best part of the Naematelia aurantialba for fruit body tissue separation,and carry out a subculture of the strains obtained from the fruit body tissue separation to determine the effective distribution range of the first generation of subculture strains Research;on the basis of the established effective strain identification technology,through the improvement of the culture medium,the determination of the mixing ratio of Naematelia aurantialba and the hairy leather bacteria,and the optimization of the preparation conditions of the Naematelia aurantialba liquid strain,etc.The identification technology and preparation method of effective strains of ears provide theoretical basis and technical support for the production of effective and high-quality strains of Naematelia aurantialba,and promote the healthy development of the Naematelia aurantialba industry.The main results of this study are as follows:1.The morphology of the Naematelia aurantialba hyphae and the hyphae of Stereum hirsutum were observed under the microscope at 40 times and 100 times.The diameter of Naematelia aurantialba hyphae was 2~5 μm,with lock-like association;the diameter of hyphae were born15~20 μm,the diameter of the Stereum hirsutum at the later stage of growth is 3-7 μm,with ring-locked association.Compared with Stereum hirsutum hyphae,the diameter of the mycelium is obviously smaller than that of Stereum hirsutum hyphae,and its toughness is poor.The hyphae are easy to break when making slices,while the hyphae of Stereum hirsutum hyphae are strong and complete under the microscope.Therefore,the size of the diameter of the hyphae and the ring-shaped linkage can be used as the basis for distinguishing the hyphae of Naematelia aurantialba from the hyphae.The hyphae of Naematelia aurantialba and the hyphae of the mixed species have no ring-locked association phenomenon,and will not show the morphological and structural characteristics of the two kinds of hyphae in a separate culture state.This finding can be used as an effective species identification in accordance with.2.The microscopic morphological observation and molecular identification of different parts of the longitudinal section of the fruit body showed that:the surface layer and sub-seed layer of the fruit body are generally distributed with aureus hyphae,from the outside to the inner to the third of the longitudinal section of the fruit body In the second place,the amount of Naematelia aurantialba hyphae is more,and the closer to the base of the ionic entity,the less the Naematelia aurantialba hyphae,but the main body of hairy bacteria;by extracting the outermost layer of the fruit body,two-thirds from the outermost layer and the DNA of different parts of the ear base was analyzed by PCR amplification,gel electrophoresis and ITS sequencing,and the results were consistent with the microscopic morphological identification results.The electrophoresis results from the outer to inner to two-thirds of the Naematelia aurantialba fruit body showed two The size of the electrophoresis band of Stereum hirsutum pilaris is 620 bp,and the size of the electrophoresis band of Naematelia aurantialba is 490 bp.Therefore,the suitable part for separation of the solid tissue of Naematelia aurantialba fungus seeds is the tissue from the outermost layer of the fruit body to two-thirds from the outermost layer.The closer to the outer epidermis,the higher the content of Naematelia aurantialba hyphae.Through the research on the culture time of the isolated strains of fruit body tissues and the effectiveness of the subculture strains,it was determined that the culture time of the effective strains of Naematelia aurantialba was more than 30 days;the strains subcultured by the female species of Naematelia aurantialba appeared If the mycelium of Naematelia aurantialba is lost,it must be identified in production before it can be used as a cultivated seed.3.Through the improvement of the culture medium and the determination of the mixing ratio of Naematelia aurantialba and Stereum hirsutum,the method for preparing effective species of Naematelia aurantialba has been studied.The research shows that adding the extract of Naematelia aurantialba to the mother seed culture medium can promote the growth of Naematelia aurantialba spores.For germination,the appropriate concentration of the extract of Stereum hirsutum edulis was selected to be 700 ml/L.The specific performance was that the spores germinated the fastest,the colony growth was good,and the hyphae growth rate was 8 mm/d;the Naematelia aurantialba yeast-like spores and hair were determined when the optimal mixing ratio of phlegm bacteria is 7:3,effective strains of Naematelia aurantialba can be prepared.4.The preparation conditions of the Naematelia aurantialba liquid strain were optimized,and the suitable medium formula for the effective strain liquid fermentation was determined as:corn flour 2%,bran 1.5%,glucose 1%,peptone 0.5%,MgSO4 0.05%,water 1000 mL and pH are natural;the optimum culture temperature is 21℃;the optimum rotation speed is 190 r/min;the optimum culture time is 8 days.
