The testis is a very important reproductive organ of male livestock.It is the place where sperm occurs and develops.Therefore,the growth and development of the testis and the reproductive activities are closely related to sperm.Multi-omics analysis provides a new method to uncover the molecular mechanism of testicular growth and development in different periods.In this experiment,RNA-seq and i TRAQ technology were used to analyze the transcriptome and proteome of pig testis to provided new ideas for revealing the molecular mechanism of testicular growth and development before and after sexual maturity in Debao pigs..In this experiment,Debao pigs of one month(3 heads)and 6 months(3 heads)were selected as the research objects,and transcriptomics and proteomics techniques were used to sequence and analyze the testes of Debao pigs.The main findings are as followed:1 Transcriptome analysis of Debao pig testisThere were 1117 genes with significant differences between the two samples before and after sexual maturity of Debao pigs,including 522 genes that were significantly up-regulated and 595 genes that were significantly down-regulated.Comparing the differential genes to GO and KEGG databases,it was found that most genes were enriched in propagation-related pathways,including: NF-κB signaling pathway,WNT signaling pathway,P53 signaling pathway,PI13k-akt signaling pathway,MAPK Signal path,etc.Through in-depth analysis of these pathways,we obtained the core genes,namely: NFATC1,FGF2,EFNA4,CD14,CCL19,CCL21,CHUK,DDX58,RPRM,APAF1,ADGRB1,COL9A2,VTN,TNN,WNT6.Finally,8 genes from 1117 genes with significant differences were selected for q RT-PCR verification,and it was found that the expression levels of these 8 genes were not different in q RT-PCR and transcriptome.It indicates that the reliability of the data obtained by transcriptome sequencing in this experiment is high.2 Proteome analysis of Debao pig testisThere were 611 proteins with significant difference between the two samples before and after sexual maturity of Debao pig,including: 347 proteins with significant upregulation and 264 proteins with significant downregulation.Comparing significantly different proteins to GO and KEGG,it was found that most of the proteins were enriched in the metabolic pathway,endocytosis pathway,heat stress pathway and reproduction pathway.A joint analysis of proteome and transcriptome found that 7446 genes related to each other in the two omics,including 188 existed in both differential genes and differential proteins.In summary,this topic has conducted a comprehensive study on the testes of Debao pigs from the transcriptome and proteome levels,and successfully screened the genes and proteins that play an important role in the growth and development of the testes of Debao pigs.It provides important data support for further in-depth exploration of the molecular mechanism of Debao pig testicle growth and development,and also provides a certain scientific basis for future Debao pig breeding.
