In this study,Gossypium up land Tm-1 and Asian gossypium subseries no.1 were used as research materials,and treated with different concentrations of EMS and Co60 with different irradiation intensities.Successive addition and character investigation and data statistics were conducted on the obtained progeny to determine different mutant types,and the obtained mutant plants were identified and analyzed.1.After continuous self-interbreeding,5 suspected mutants were finally obtained in the mutated cotton,namely villi mutant,plant mutant,brown cotton mutant,green cotton mutant and premature aging mutant.2.The obtained mutant materials were identified.Through in vitro culture experiments,identification of pigment content in leaves,detection of senescence related enzyme activity in leaves,determination of senescence related gene expression and investigation of agronomic traits,the results showed that the obtained mutant was of the senescence type.3.The main characteristics of the progeria mutants were early flowering and accelerated aging,but there was no significant difference between the wild type and other agronomic traits of cotton,including seed value,lint fraction and boll number.4.Sampling wild-type and premature aging mutants in different periods for transcriptome sequencing,obtained 161672588 raw reads,Q20 at 98.04%-98.13%,Q30 at98.02%-98.10%,GC content at 44.14%-44.38%.After filtering low-quality reads,158374438 clean reads were obtained.A total of 17,868 expressed genes were detected;GO functional annotation showed that differentially expressed genes were enriched into 3major categories and 30 subcategories;differential gene KEGG annotation analysis showed that differentially expressed genes were mainly distributed in the photosynthesis-antenna protein pathway,Nitrogen metabolism,plant pathogen interaction,phytohormone signal transduction,starch and sucrose metabolism,and flavonoid biosynthesis pathways suggest that these processes may play an important role in the premature aging of upland cotton.5.A large number of mutation sites were found in the premature aging mutation pool,which can occur in all regions of the gene,most of which are distributed in Exon and Intro regions.Most of the variation sites were Missense and Slient,while a few were Nonsense.From the perspective of Modifier,Modifier was the most common Modifier,followed by Low and Moderate,while High was the least common Modifier. |
