In order to research the changes of phosphatase activity of soil in apple-pear orchard under different seasons,different planting years,and different soil layers,the effect of seasonal freezing and thawing on phosphatase activity,and the synergistic mechanism between phosphatase activity and phosphorus,the soil of apple-pear orchards in Yanbian was selected from different orchards of different planting years,soil samples were collected and processed according to different soil layers and years of planting.The modified phenylphosphonate method and p-nitrophenyl phosphate disodium method were used to determine the different phosphatase activities in the soil,and the Bray method for determination of available phosphorus,improved Bowman-Cole method by Xiong-Fan for grading the soil organic phosphorus,analyzing and researching the effects of season,planting years,soil layer and freeze-thaw on phosphatase,the relationship between soil phosphatase,organic phosphorus components and available phosphorus of soil in apple-pear orchard.The results showed:1.Phosphatase activity in soil:acidic>neutral>alkaline.Phosphomonoesterase activity:acidic>alkaline.The acidic and neutral phosphatase activities in autumn were significantly greater than that in spring and summer.Alkaline phosphatase was basically not affected by the season.Phosphomonoesterase activity gradually increased from spring to autumn.Phosphodiesterase activity in autumn was significantly greater than that in spring and summer.Phosphatase and Phosphomonoesterase activities first decreased and then increased with the growth of planting years.The activity of phosphodiesterase of soil in 10-year-old orchard was significantly greater than that of 25,40 and 60 years old.The activities of phosphatase,phosphomonoesterase and phosphodiesterase decreased with the deepening of soil layers.2.After seasonal freez-thaw,the phosphatase activity of the orchard soil planted for 10 years was significantly enhanced,and other changes were not significant.Seasonal freeze-thaw significantly affected the activity of acid phosphomonoesterase.The acid phosphomonoesterase activity decreased at 10,25,and 40 years old orchard,and increased significantly at the 60-year-old orchard.It had little effect on activity of alkaline phosphomonoesterase,and the soil layer of 40-60 cm was basically unchanged except for the 10-year-old.Seasonal freez-thaw promoted the reduction of phosphodiesterase activity,which had a significant impact on the 25-year-old orchard.3.The activity of soil acid phosphatase was positively corre lated with the content of available phosphorus,labile organic phosphorus,moderately labile organic phosphorus and total extracted organic phosphorus.The activity of neutral phosphatase was positively correlated with the content of available phosphorus.Alkaline phosphatase activity was negatively correlated with the content of highly stable organic phosphorus.The activity of acid phosphomonoesterase was positively correlated with the content of available phosphorus and highly stable organic phosphorus.Alkaline phosphomonoesterase activity was positively correlated with available phosphorus content,Alkaline phosphomonoesterase activity was negatively correlated with highly stable organic phosphorus content.Phosphodiesterase activity is negatively correlated with highly stable organic phosphorusMonitoring and evaluating soil fertility quality and soil health status through the use of soil biological activity,providing a basis for improving the utilization of phosphorus in economic crops and promoting yield is an important direction of future soil science research. |
