| Halofuginone hydrobromide is one kind of salts of halofuginone,and has efficient effect on many types of coccidiosis.Halofuginone hydrobromide is widely used as a kind of additive in feeds for prevention and treatment of chicken coccidiosis.In order to judge the effect of the medicine on growth,the feeding experiment was carried out.However,during the long-term feeding,chicken were very sensitive to concentration of the drug,which could cause toxic effect by high content or uselessness by lower concentration.Therefore,there is an urgent demand for monitor of halofuginone hydrobromide in feed.As described in this paper,a HPLC method was established to detect the compound in feed.This experiment also chicken blood detection method is established,to study the pharmacokinetic process and calculate the bioavailability of halofuginone hydrobromide.Results are listed in detail as follows:1 Development of RP-HPLC method for determination of halofuginone hydrobromide in feedsA solid-phase extraction and reversed-phase high performance liquid chromatography(SPE-RP HPLC)method for determination of halofuginone hydrobromide in feeds was developed in this study.Halofuginone hydrobromide in feeds was successively extracted by ethyl acetate and 0.125 mol·L-1 ammonium acetate buffer,and was cleaned up via HLB column.The mobile phase was consisted of acetonitrile,0.25 mol·L-1 of ammonium acetate buffer and water(5:3:12-V/V/V),and adjusted to pH value equal to 4.3 with glacial acetic acid.Halofuginone hydrobromide was monitored by UV detector at 243 nm.The result showed that,the limit of detection and quantification was 0.05 μg·g-1 and 0.2 μg·g-1,respectively.Among the concentration range from 0.2 to 15 μg·g-1,the correlation coefficient is greater than 0.999,a good linear relation.In 0.2,1.5,15 μg·g-1 levels,the rate recoveries of 87%~113%was obtained,and within-day and between-day cofficents of variation were 1.1%~17%and 7.1%~12%respectively.The research results show that this method can meet the requirements of sensitivity,precision and accuracy,which can be applied for quick and efficient determination of halofuginone hydrobromide in poultry feeds.2 Study on the pharmacokinetics and bioavailability of halofuginone hydrobromide in BroilerTo study the pharmacokinetic process and calculate the bioavailability of halofuginone hydrobromide in healthy chickens body this study chooses 36 healthy chickens to be randomly grouped into 2 equal-sized groups.A single dose of hydrogen bromide premix plus 0.6 mg·kg-1·bw and intravenous halofuginone hydrobromide is the one that is 0.3 mg·kg-1·bw then the concentration of halofuginone hydrobromide is determined by using UPLC-MS/MS.The pharmacokinetic parameter is calculated with 3p97 program software processing Blood concentration-time data.Statistics found that broiler non-absorbent after oral halofuginone hydrobromide premix,and different animals have different differences,all the chicken has the highest drug concentration in the body at about 8 ng·mL-1.Most of the chicken’s blood is still below the quantitative limit in 8h and the 72 h is basically eliminated below the quantitative limit.The result indicates that after injection taking halofuginone hydrobromide,here are the main pharmacokinetic parameters:T1/2β(23.66±13.5)h,CL(0.95±0.32)·L·h-1·kg-1,Vd(29.23±13.72)L·kg-1,AUC0-∞(361.02±152.19)ng·h·mL-1,MRT0-∞(34.15±19.48)h.The results showed that the it was rarely absorbed after oral halofuginone hydrobromide premix by the intestina.3 The effect of the premixture of halofuginone hydrobromide on the growth ofthe chickenTo study the clinical effect of halofuginone hydrobromide premix as an additive,600 Partridge-shank Broilers were randomly and averagely divided into five groups:high-dose treated group,medium-dose treated group,low-dose treated group,control group,and Maduramicin ammonium treated group.Animals were fed for under 42 days under usual feeding situation.Deaths and clinical symptoms were recorded at 0,14,28,and 42 days respectively.Data were analyzed by SPSS.As a result,mortality rate of high-dose treated group and medium-dose treated group was significantly higher than Maduramicin ammonium treated group,moreover,the high-dose group was very significantly higher than control group.At the last stage,the average weight of the high-dose treated group was lightest,and the feed intake was decreased heavily.Therefore,used as additive.halofuginone hydrobromide had not exhibited significant effect on chicken growth,while long-term challenge of high doses might cause poor mental state,decreased feeding and weight gain,and high mortality. |
