Pine wood nematode(North American pinewood nematode;Bursaphelenchus xylophilus)and Bursaphelenchus mucronatus(Bursaphelenchus mucronatus)belong to the same linear animal door,sliding blade head(Aphelenchida),sliding blade suborder(Aphelenchina),sliding blade superfamily(Aphelenchoidoidea),sliding blade section(Aphelenchoididae),Bursaphelenchus(Bursaphelenchus).The pine wilt disease caused by Bursaphelenchus xylophilus was known as the pine" AIDS",at present mainly in the United States,Canada,Spain,Mexico,Portugal,China,Japan,Korea and other 8 countries,the most serious is China,Japan and South Korea belong to the international important quarantine objects of forest diseases and pests,listed as the first.This research mainly to pine wood nematode,Bursaphelenchus mucronatus as materials,their genetic polymorphism were compared and the construction of nematode fingerprint.Obtained the following results:(1)The introns cut contact primers in the PCR molecular markers,of 14 copies and 3 copies of pine wood nematode Bursaphelenchus mucronatus and 1c.elegance genetic diversity were studied,the results show that:by using 9primers were in the 18 materials stable amplification striping,received a total of 189bands polymorphic bands,including140,the average ratio of polymorphism of 74.07%,average every pair of primers amplified 15polymorphic bands.According to the ISJ molecular markers in 18nematode materials reveal genetic distance,using a weighted average method,UPGMA method genetic cluster analysis,18nematodes material can be clearly divided into pine wood nematode Bursaphelenchus mucronatus and2 categories.And also reveals the interspecific genetic distances were significantly greater than intraspecific genetic distance.(2)Nematode DNA fingerprinting constuction:in accordance with the standard DNA fingerprint database construction principle,selected from3 primers(E4,Rland R4),16 polymorphic loci,will each strip with or without were recorded as 1 or 0 method,used to construct 18 nematode material DNA fingerprints database,so that in this nematode DNA fingerprint database,each nematode materials have specific DNA fingerprint.At the same time according to each point of amplified band has no,the construction of 18 C material DNA fingerprints into the computer can identify the digital fingerprint table,so that each a nematode material with a specific code sequence corresponding to.Study design were prepared with a database and digital recognition in all 2modules of the nematode digital fingerprint identification software-pngl.In the coding of digital identification module,input a fingerprint code,the software can automatically and the fingerprint database of species in the digital fingerprint matching.This work for the automated identification of nematode species and species resources database construction and development of the foundation,but also in the invasive plant species in rapid quarantine,also have potential application value.In order to improve breeding efficiency.and select fine barley germplasm resources,7 different wild barley populations from Israel and Jordan with 133 lines of agronomic traits was studied and analysised,the results show that,7 populations with plant height,spike length,awn length,flag leaf length,peduncle and extrusion,grain number per spike,panicle number,flag leaf width,number of tillers have significant difference within and between populations and have abundant genetic diversities;The characters of variation coefficient is high and the most notable character is the number of panicles per plant,the range of variation is 0.19-0.34,It could provide abundant genetic basis of crop breeding;133 experimental materials of agronomic traits in accordance with the average UPGMA was used to cluster analysis,The clustering results can be divided into 7 classes,better reflect each class agronomic characteristics,advantages and disadvantages in each class and can be complementary;According to the different breeding objectives and focusing on the selection of the desired traits,to improve breeding efficiency.
