This thesis comprehensively explored the anti-colon cancer effects of luteolin in and out of the body and the relationship between luteolin and p53 signaling pathway.The research contents included exploring the effects of luteolin on proliferation,metastasis,cycle and apoptosis of colon cancer HT-29 cells,and exploring the relationship between luteolin and p53 signaling in colon cancer HT-29 cells at the molecular and protein levels.To explore its possible anticancer mechanism.The effect of luteolin on the proliferation of colon cancer HT-29 cells was investigated by CCK8 cytotoxicity assay.The inhibitory effect of luteolin on HT-29 cell migration was evaluated by cell scratch assay and Transwell invasion assay.The effects of luteolin on cell cycle and apoptosis of HT-29 cells were evaluated by flow cytometry.Changes of p53 signaling pathway were detected at gene and protein levels by RT-PCR and Western-blot assay.A nude mouse model of colon cancer was established,the changes of body weight,blood sugar and other physiological indexes were monitored,and the intracellular anti-tumor effect of luteolin was further evaluated by slice HE staining.It was found that the growth and proliferation ability of colon cancer HT-29 cells were significantly inhibited by luteolin after 24 h,48h and 72 h of different concentrations of luteolin in vitro,and the inhibitory rate of luteolin on colon cancer cells showed a significant concentration and time dependence.In migration and invasion experiments,luteolin also significantly inhibited HT-29 migration and invasion.The results of flow cytometry showed that luteolin could effectively block HT-29 cells in S phase and induce apoptosis significantly.Western blot test showed that luteolin up-regulated the proportion of p53,p21 and p27 proteins,and RT-PCR test confirmed that the expression levels of p53,p21 and p27 genes increased,and showed a concentration dependence with luteolin.On this basis,a nude mouse model of colon cancer was established.The changes in body weight and blood glucose of nude mice after administration suggested that luteolin was safe,and the changes in tumor volume after administration and the results of HE staining showed that luteolin showed a significant anti-tumor effect in vivo.In summary,luteolin can inhibit the proliferation and migration of HT-29 cytotoxicity and block cells in the S phase,and its molecular mechanism may be related to the regulation of p53 signaling pathway,and promote the apoptosis of HT-29 cells by blocking the cell cycle.This study provides a new research idea for the subsequent clinical application of luteolin in the treatment of colon cancer.
