Cudrania tricuspidata is a kind of Cudracula plant of mulberry family.Its leaves,fruits,bark,roots,trunks and other different parts have been studied by scholars.Research reports that the extracts from different parts of Cudrania tricuspidata have antioxidant,anti-tumor,bacteriostatic and other effects.In this paper,Cudrania tricuspidata leaf was selected as the research object,and ultrasonic assisted water bath was used to extract flavonoids and polysaccharides from Cudrania tricuspidata leaf.After separation and purification,the structure was analyzed,finally application in cosmetics.The specific research results are as follows:In this paper,ultrasonic assisted water bath was used to extract flavonoids from Cudrania tricuspidata leaf.Through single factor experiment,the extraction conditions of flavonoids were as follows:ethanol volume fraction 50%,material liquid ratio 1:40,time 30 min,temperature 70℃.After TLC purification,UV and IR spectra analysis showed that there was an absorption peak at 271.1 nm in the UV spectrum of the extract of Cudrania tricuspidata leaf,which could be used to determine that the sample might contain benzene ring;There is a strong absorption peak near 1640.1 cm-1 in the infrared spectrum,which can judge that the sample may contain C=O double bond.All the above are characteristic absorption of flavonoids,so it is determined that the extract is flavonoids.In this paper,ultrasonic assisted water bath extraction of CTLP was used.Through single factor combined with orthogonal experiment,the extraction process of polysaccharide from Cudrania tricuspidata leaves was optimized,and the optimal extraction process parameters of CTLP were obtained as follows:temperature 70℃,time 60 min,material to liquid ratio 1:40.Under this condition,the sugar content of CTLP was 6.22%.High speed countercurrent chromatography(HSCCC))was used for separation and purification.A solvent system of 12%PEG800-15%anhydrous Na2CO3-73%H2O(w/w/w)was selected.Finally,a refined polysaccharide CTLP-1 from Cudrania tricuspidata leaves was obtained.Preliminary structural identification of CTLP-1.The content of nucleic acid and protein was determined by UV,the molecular weight was determined by HPGPC,the composition of monosaccharide was determined by IC,and the polysaccharide skeleton was determined by IR.The results showed that CTLP-1 contained no nucleic acid and protein;It contains three components with different number average molecular weight(192515 Da,28031 Da,6357 Da),and the proportions of each component are 14.838%,80.3215%,4.841%respectively;Six kinds of monosaccharides including Rha,Ara,Glc N,Gal,Glc,Gal A,with the molar ratio of 0.027:0.183:0.002:0.255:0.524:0.010;It contains the basic skeleton of polysaccharides and functional groups such as O-H,C-H and C=O.The ability of scavenging DPPH free radical and hydroxyl free radical was determined.Two skin care products with different substrates,namely skin care water and moisturizing milk,were designed.The stability and moisture retention of the water emulsion samples were determined.The results showed that both CTLP and CTLP-1 had antioxidant activity;Polysaccharide added water emulsion has good physical and chemical properties,good stability and excellent moisture retention.
