Emulsion is the basis of many foods,cosmetics and pharmaceuticals.Stimulation response emulsion can rapid regulation between steady state and unstable state,and it is the delivery carrier of drugs and functional components with highly maneuverable and broad application.Glycerides is a mixture composed of triglyceride,diglycerol and monoglyceride.The structural esters are the basis of emulsifying properties,stability and good biological activity of glycerides.Thus,the emulsion composed of glycerides has great development potential.High-methoxyl pectin is plant polysaccharides,it has excellent emulsifying properties and stimulus response characteristics of pH,which makes it widely used in biomedical and food industries.Thus,in this research,high pressure homogenization method was used to optimize the preparation of CG-HMP which with high-methoxyl pectin as wall material and glycerides as core material.And we studied the physicochemical properties,characteristics of in vitro digestion and azithromycin delivery of CG-HMP.It is expected to provide a theoretical basis for the application of oil-in-water emulsion in drug delivery.1.Optimization of preparation of CG-HMPThe high-pressure homogenization method was used to prepare CG-HMP.The embedding rate is taken as the comprehensive index.The single factor test,Plackett-Burman design,steepest climb test and response surface analysis were used to optimize the preparation conditions of CG-HMP.Optimum preparation conditions of CG-HMP were as follows: wall material concentration of 3.50%,oil phase content of 14.39%,emulsifier content of 1.50%,homogenizing pressure of 60 MPa,homogenizing cycle 4 times.2.Physicochemical properties of CG-HMPThe emulsion type,particle size and potential,infrared characteristic,apparent property and stability were studied.The results showed that the type of CG-HMP was oil-in-water,the average particle size of CG-HMP is 480.47±76.19 nm,and CG-HMP has negatively charged,an obvious putamen structure and high embedding rate.CG-HMP has good stability in strong acid environment(pH2.0-4.0)and low temperature environment(5 ℃).3.In vitro digestion characteristics and application of azithromycin delivery of CG-HMPThe digestive properties and drug delivery applications of CG-HMP were studied by the method of in vitro simulation.The results showed that CG-HMP could significantly reduce the production of free fatty acids during digestion;and CG-HMP had good stability in the oral and stomach,and could rapidly demulsify in the small intestine part.CG-HMP had a certain stimulus response characteristic of pH.The release properties of azithromycin in CG-HMP were in agreement with the first-order drug release equation,and it showed a sustained release effect and an obvious targeting effect of small intestine.These could effectively reduce the occurrence of gastric adverse reactions of azithromycin.The results showed that CG-HMP is a good targeted delivery system of drug.
