Allylsilane is an environmentally friendly allylation reagent.Under Lewis acid or transition metal catalyst,it can react with numerous substrate molecules to introduce allyl functional groups.thus having important applications in organic synthesis.In the synthesis of semiterpene natural products and drug molecules.In addition,the allylation reaction is often a critical step.Allylsilane also shows very good reactivity and high selectivity as a mild reagent.Although studies on allylsilanes have provided many effective strategies.Preparation of highly stereoselective allylsilanes remains a challenge.Especially,the synthesis of functionalized allylsilanes.Compatibility issue has not been well addressed.In this thesis,an indepth study of functionalized allylsilane reagents was carried out and applied to the construction of in silico drug molecular backbones with potential drug activity.The main scientific findings are as follows.Efficient synthesis of complex organosilicon containing molecules using free radical reactions has been noticed.But,the application to the synthesis of functionalized allylsilanes has not been reported in the literature.In this thesis,a series of functionalized allylsilanes were prepared based on a visible-light-catalyzed approach using easily prepared Morita-Baylis-Hillman(MBH)acetate and silylic acid as raw materials,and the reaction was characterized by mild conditions,easy operation,good substrate universality,and high stereoselectivity(>20:1 Z/E).1,3-dihydroisobenzofurans are one of the important drug backbones.In this thesis,a series of intramolecularly functionalized allylsilanes were designed and synthesized.Under Lewis acid catalyst,a series of silylated 1,3-dihydroisobenzofuran compounds with potential physiological activity were efficiently synthesized by Sakurai allylation reaction.This work is of great value in providing a new route with practical value for the development of related silyl derivatives.
