Corn silk is composed of various chemical components such as polysaccharides,flavonoids,saponins,alkaloids,and organic acids.These components have been found to possess medicinal properties such as anticancer,hypoglycemic,diuretic,and antifatigue effects.While some studies have shown that corn silk may also affect coagulation activity,further research is necessary to fully understand its mechanism of action.This paper presents a systematic screening of the procoagulant active sites of corn silk through in vitro coagulation experiments.Additionally,an optimal extraction process was identified for the effective sites of corn silk based on hypoglycemic activity.Using these findings,a corn silk plant beverage was prepared,providing a scientific basis for further research and development.The study methods and main results are outlined below:1.Screening and validation of effective coagulation components in corn silk: Sequential extraction and purification of polysaccharides,flavonoids,and saponins from corn silk were performed,and their effects on coagulation were evaluated using APTT,PT,TT,and FIB as judgment criteria.Tranexamic acid,heparin sodium,and 0.9% saline were used as positive,negative,and blank controls,respectively,to screen for coagulation active sites.The results showed that the coagulation activity of corn silk polysaccharides had a dose-dependent relationship,and at 500 μg/m L,the PT and APTT values were significantly shortened compared to the blank control group(P≤0.001)and was close to the positive control group tranexamic acid.The coagulation time PT,TT,and APTT values of corn silk flavonoids and saponins were significantly prolonged(P ≤ 0.001).After the purification and separation of corn silk polysaccharides,the coagulation time PT,TT,and APTT values of the five components were significantly shortened(P ≤ 0.001),further verifying the procoagulant activity of corn silk polysaccharides.2.Spectrum-effect relationship of coagulation activity of polysaccharides from corn silk from different origins: The polysaccharides from Anhui,Henan,Yunnan,Sichuan,Sichuan,Jilin,Heilongjiang,Hebei,Guangxi,Shandong and Gansu provinces were extracted and purified,their coagulation activity was determined,and the spectral relationship of polysaccharides from different regions was established.The degree to which corn silk polysaccharide contributed to the efficacy was initially determined.Corn silk polysaccharides exhibited procoagulant activity on APTT,PT,and TT,as well as the three coagulation pathways.Spectral effect relationship analysis and orthogonal partial least squares(OPLS)regression model indicate that corn silk polysaccharides have the effect of promoting exogenous coagulation factors and promoting thrombin or fibrinogen,among which glucose,xylose,arabinose and isolated monosaccharides have the same main components of procoagulant activity.Among them glucose,xylose and arabinose were consistent with the main components responsible for the procoagulant activity of the isolated monosaccharides.3.Preparation of corn silk water extract and its product based on hypoglycemic activity:The corn silk water extraction process was optimized using response surface methodology and analytic hierarchy process.The optimal conditions were determined to be 3 g of corn silk,extraction time of 50 minutes,liquid-to-material ratio of 35:1 m L/g,and extraction for 3 times,resulting in a polysaccharide extraction rate of 3.88% and a flavonoid extraction rate of 1.57%,the combined filtrate was concentrated to 7 m L,the inhibition rate of α-glucosidase was as high as 79.16%.The Lineweaver-Burk double reciprocal curve analysis showed that corn silk had a mixed type of reversible competitive and noncompetitive inhibition on α-glucosidase.4.Research on corn silk plant beverage: Based on the above-mentioned process,a plant beverage with sucralose,sodium isoascorbate,xanthan gum,cyclodextrin,food flavor as excipients,and the optimal mouthfeel was obtained by sensory scoring combined with single factor experiments,the beverage was subjected to nutritional component detection and microbiological detection,and the results all met the national standard requirements.Finally,the safety of corn silk beverages was evaluated at a 5-HMF level of 0,and hemolysis of red blood cells was not induced in these experiments.Therefore,the quality and safety of the product are demonstrated.In this paper,from screening the effective sites of blood coagulation,exploring the components and Spectrum-Effect relationships of blood coagulation,the method of effective site extraction,and the study of products,a systematic approach from basic research to applied research,provides a deep scientific basis for developing new medicinal value and resource utilization of corn silk.
