With the improvement of living standard,the number of hypertensive patients in China is increasing day by day.Most of the usual treatment methods are to use drugs to regulate blood pressure,but this will produce many side effects,so it is necessary to develop microecological agents with blood pressure regulation effect.Therefore,in this paper,Lactobacillus plantarum 2-33,which has been evaluated for safety and has an inhibitory effect on ACE,was combined with Lactobacillus plantarum 43 and Lactobacillus plantarum10,which produce γ-aminobutyric acid,to develop a compound microecological preparation and used in the preparation of fermented milk to study its inhibitory effect on ACE.The results of the study were as follows:1.By evaluating the safety of 2 strains of Lactobacillus plantarum(43 and 10),the results of the hemolysis test,antibiotic susceptibility test and oral acute toxicity test in mice showed that the 2 strains of Lactobacillus plantarum were safe for consumption.2,three strains of Lactobacillus plantarum(2-33,43,10)were compounded in a 1:1:1ratio(mass ratio)to determine their optimal growth temperature of 37°C.The optimal formulation of the lyophilized protective agent of the composite microecological formulation was determined by single-factor and response surface experiments as 11.70%skim milk,5.60% monosodium glutamate and 2.66% Tween-80.Under these conditions the live count of the composite microecological preparation was 5.32×10~9 CFU/g with the highest survival rate of 88.70%.3.The results of pH 2.0 tolerance test,bile salt tolerance test and simulated artificial gastric juice tolerance test showed that the compound microecological preparation was strongly tolerated under pH 2.0 and 0.30% bile salt environment,and it could pass the gastrointestinal barrier smoothly and exert probiotic effects in the body.The viable bacteria count could still reach 6.6×10~8 CFU/g during the storage period of 90 d.Therefore,the compound microecological preparation has good storage ability.4.The acidity of the fermented milk prepared with the compound microecological preparation was suitable during the storage period of 28 d(titratable acidity of 89.30°T);the viscosity showed a trend of increasing and then decreasing,reaching the highest value(1322mpa·s)at the 14 th day;it indicated that the fermented milk had a stable structure and smooth texture.The maximum number of viable bacteria was 8.15 lg(CFU/m L)at the 7th day.By measuring the hygienic indexes,we learned that the coliform count of fermented milk was≤5 CFU/m L,and no yeast,mold and Staphylococcus aureus were detected,meeting the requirements of the Food Safety National Standard for Fermented Milk for fermented milk.5.The inhibition rate of fermented milk supernatant on ACE was 66.50% as measured by high performance liquid chromatography,so the fermented milk prepared by the compound microecological preparation had good inhibition effect on ACE.Therefore,a complex microecological preparation developed by three strains of Lactobacillus plantarum(2-33,43,10)that can be applied to fermented milk production and has an inhibitory effect on ACE has an important application value.
