| Blumea balsamifera(L.)DC.is a perennial herb or subshrub of the Asteraceae family,a traditional Chinese Miao medicine with antiseptic and anti-inflammatory,antioxidant and analgesic effects,and its main application form is the essential oil extracted from Blumea balsamifera(L.)DC..However,due to the insolubility and high volatility of the Blumea balsamifera(L.)DC.essential oil(BBO),it is often difficult to promote the application of the oil because of its short duration of efficacy and uncontrollable dosage.In order to improve the scope of medicinal use of the Blumea balsamifera(L.)DC.essential oil,the preparation of the oil preparations was started.Blumea balsamifera(L.)DC.essential oil has been used in folk medicine for wound healing,but the mechanism underlying its effect on skin wound healing has not been investigated.In this paper,we investigate the effect of Blumea balsamifera(L.)DC.essential oil on skin wound healing and the related mechanism by combining in vivo and in vitro experiments,in order to provide some data support for the promotion of the application of Blumea balsamifera(L.)DC.essential oil in skin wound healing.The main findings of this paper are as follows:1.Blumea balsamifera(L.)DC.essential oil gel was prepared and its skin wound repair effect was evaluated.Compared with the blank group,the wound repair rate of Blumea balsamifera(L.)DC.essential oil gel increased by 26.4%(P<0.05),indicating that Blumea balsamifera(L.)DC.essential oil gell has the ability to heal skin wounds.2.A nanoemulsion of Blumea balsamifera(L.)DC.essential oil(BBO-NE)was prepared and its skin wound repair effect and mechanism were discussed.After 7 days of application,the nanoemulsion form significantly increased the efficacy of Blumea balsamifera(L.)DC.essential oil for wound repair.The number of inflammatory cells was significantly lower(P<0.05)and collagen deposition was significantly higher(P<0.05)in the nano-formulation group compared to the Blumea balsamifera(L.)DC.essential oil group.In addition,inflammation-related TLR4,CD14 and IRAK-1 gene transcript levels were significantly reduced after the administration of BBO-NE compared to the BBO group,with downregulation of 47.8%,35.7% and 57.8%,respectively,while the secretion of pro-inflammatory factors IL-6 and TNF-α was also significantly reduced by 83.8% and 32.7%,respectively,in the nanoformulation administration(BBO-NE)group compared to the BBO group.In contrast,the anti-inflammatory factor IL-10 was significantly increased by 4.2-fold.The nano dosage form increased the skin permeability of Blumea balsamifera(L.)DC.essential oil by 6.3% to 19.5%(P<0.05),and improved the drug bioavailability.Conclusion: Blumea balsamifera(L.)DC.essential oil has trauma healing effects,and the nano-emulsion improved the skin absorption of Blumea balsamifera(L.)DC.essential oil,exacerbated the down-regulation of inflammation-related genes TLR4,CD14 and IRAK-1 transcript levels,significantly reduced inflammatory factor production,increased anti-inflammatory factor levels and promoted collagen deposition,which in turn enhanced the trauma healing effects of Blumea balsamifera(L.)DC.essential oil. |
