| Infections caused by foodborne pathogens pose a serious threat to human health,affecting hundreds of millions of people in both developed and developing countries each year.E.coli is one of the most common foodborne pathogenic bacteria,and E.coli O157:H7 is a representative and most harmful type of E.coli.E.coli O157:H7 is mainly transmitted through unclean water and food.Therefore,there is an urgent need to establish a detection method for E.coli O157:H7that is easy to operate,sensitive and time-consuming,which is of great significance for monitoring food safety and human health.In this study,two electrochemical detection methods of E.coli O157:H7 were established based on screen-printed electrodes and lateral flow immunochromatography strips.Study 1.Electrochemical detection method of E.coli O157:H7 based on antibody and anti microbial peptide.In this detection method,the screen printing electrode was fixed under the lat eral flow immunochromatography test strip,the E.coli O157:H7 specific antibody was fixed on the test strip detection line,the E.coli O157:H7 antibacterial peptide was coupled with ferrocen e,and the antibody-E.coli O157:H7-antibacterial peptide formed a sandwich structure.Under o ptimal conditions,the linear range of E.coli O157:H7 was 1×10~3CFU/m L~1×10~8CFU/m L,the linear equation was y=0.2620Lg C-0.7162,and the detection limit was 2.56×10~2CFU/m L.The e xperimental results show that the experimental detection method has good specificity and high s ensitivity,and the whole detection process takes 30 minutes,which has good applicability.Study 2.Establishment of electrochemical detection method for E.coli O157:H7 based on antibody nanoflower and antibacterial peptide.In this assay,E.coli O157:H7 was used as the experimental target.The screen printing electrode was fixed under the lateral flow immunochromatography test strip,and the E.coli O157:H7 specific antibody nanoflower was fixed in the detection line to detect E.coli O157:H7 antibacterial peptide coupled with ferrocenecarboxylic acid,and the antibody nanoflower-E.coli O157:H7-antibacterial peptide formed a sandwich structure.Under optimal conditions,the linear range of E.coli O157:H7 was1×10~2CFU/m L~1×10~8CFU/m L,the linear equation was y=0.3158Lg C-0.4950,and the detection limit was 2.5×10~1CFU/m L.This test can be completed in less than 20 minutes,which is shorter than traditional E.coli detection methods,highly sensitive and can achieve visual readings of test results.It is important for rapid and sensitive visual detection of foodborne pathogens. |
