| Hydroxyapatite(HA)has good biocompatibility,biological activity,but also has a certain bone induction,as one of the most important inorganic components of human bone tissue,hydroxyapatite has been widely concerned and applied in bone tissue repair and bone defect filling process.Although nano-hydroxyapatite powder itself has good biocompatibility,but also has a certain cytotoxicity.To avoid the above problems,this paper through the microwave hydrothermal method efficiently a large amount of amino acid was prepared by modified nano-hydroxyapatite powder,powder made by small grain size,length-diameter ratio is low,very uniform,the characteristics of morphology and cell toxicity is small,then doxorubicin(DOX)load on hydroxyapatite powders,found that has a certain inhibitory effect on the osteosarcoma cells.(1)The effects of p H and different amino acid ratio on the hydroxyapatite powder were studied by microwave hydrothermal method.The results showed that the nanohydroxyapatite powder was rod-like when the reaction temperature was 160 ℃ and the reaction time was 40 min.The length and diameter of nano-hydroxyapatite particles were 64.7±13.6nm and 29.1±3.9nm when the mole ratio of amino acid to calcium ion was 1.5.(2)Different amino acid feeding ratio has an effect on the grain size and dispersion of hydroxyapatite,so the specific surface area of hydroxyapatite powder modified by different concentrations of amino acids is different.The prepared nano-hydroxyapatite powder is used as drug carrier to load doxorubicin(DOX).The drug loading capacity of hydroxyapatite powder modified by different amino acids and its inhibitory effect on osteosarcoma cells were studied.Hydroxyapatite powder modified with threonine can inhibit osteosarcoma by loading doxorubicin.(3)Nano-hydroxyapatite powder modified with different amino acids was prepared by microwave hydrothermal method.Through morphology analysis and in vitro cell experiment,it was found that hydroxyapatite powder modified by threonine had less toxicity to mouse osteoblasts,and had obvious inhibitory effect on osteosarcoma cells. |
