The Research Of The Biological Efficacy Of DDW And The Extraction Of Main Ingredients From Tea And Chinese Medicine With DDW

Water is an important substance that sustains life and it is a compound composed of hydrogen and oxygen atoms.The hydrogen has three isotopes which are hydrogen（protium,H）,deuterium（D）and tritium（T）.The ratio of deuterium to hydrogen（D/H）in ordinary water is about 1:6600 which means that the volume fraction of deuterium in water is 0.015%.Water with a deuterium volume fraction of less than 0.015%（150 ppm）is generally referred to as deuterium depleted water（DDW）.In this thesis,DDW of 40,70 and 100 ppm were used as the study subjects,and distilled water was used as the control group,The effects of DDW on the four blood lipids,immunity,intestinal flora and the pre-protective effect on mice with liver damage were studied at the animal level,the inhibitory effect of DDW on cancer cells was studied at the cellular level and its effect on the extraction rate of main ingredients from tea and chinese herbs.The key findings are as follows:4.Efficacy studies at animal level:the content of deuterium in serum and liver of mice decreased to some extent after drinking DDW,and the lower the deuterium content in water,the greater the reduction in serum and liver deuterium levels.In the control group the deuterium content in the liver of mice was less than that in the serum,and after drinking DDW the deuterium content in the liver of mice was greater than that in the serum,and the formation of the opposite isotopic gradient may increase physical resistance.DDW reduced the content of total cholesterol（TC）and low-density lipoprotein cholesterol（LDL-C）;DDW increased the thymus index in mice,which may have the effect of improving immunity.DDW has beneficial effects on the body’s health by regulating the intestinal flora.It mainly showed that the DDW reduced the relative abundance of firmicutes and increased the relative abundance of bacteroidetes.DDW decreased the relative abundance of barnesiella and increased the relative abundance of prevotella,and prevoella plays a key role in carbohydrate metabolism.DDW decreased the activity of ALT and AST in serum,decreased the contect of MDA in liver tissue,and increased the activity of SOD in liver tissue,the preprotective effect of DDW on acute liver injury induced by CCl₄in mice may be due to the enhancement of antioxidant capacity of the body to reduce liver damage.1.Research on the efficacy of DDW at the cellular level:DDW inhibits the proliferation of Hela,MCF7,ES-2,HT-29,and HepG2 cells,the lower the deuterium content the more obvious the inhibitory effect,while DDW does not affect the growth of normal cells;DDW interferes with HepG2 cell cycle,which mainly shows that DDW increases G1 phase and decreases S phase,It is hypothesized that DDW may inhibit the proliferation of cancer cells by blocking the transition from the G1 to the S phase.2.DDW improved the leaching rate of caffeine and tea polyphenols in different teas,and the stability of Lapsang Souchong tea soup brewed with DDW was better.DDW facilitated the extraction of saponin components and improved the extraction rates of total saponins from Polygalae Radix,Ziziphi Spinosae Semen,Astragali Radix and Bupleuri Radix and also improved the extraction rate of polysaccharide from Codonopsis Radix and alkaloids from Sophorae Flavescentis Radix,the lower the deuterium content,the higher the extraction rate.
