| Ganpu tea is a kind of compound tea made from Pu’er tea and tangerine peel by baking and aging.Pu’er tea and tangerine peel are rich in polyphenols,such as tea polyphenols and flavonoids,which can resist oxidation,reduce blood lipid,prevent and improve obesity.Obesity is a metabolic disease caused by the accumulation of energy into fat due to the excessive intake of energy,which is closely related to diet.When taken orally,the polyphenols in Pu-erh tea and tangerine peel can modulate the composition of intestinal microorganisms,influence related metabolites such as the production of short-chain fatty acids in feces and other related metabolic markers,increase the content of beneficial bacteria in intestinal microorganisms,reduce harmful bacteria,prevent obesity and improve the phenotype of obesity.The interaction between tea and intestinal microorganisms may explain the role of tea in preventing and improving obesity to some extent.Ganpu tea appears in people’s daily life as a health beverage.In order to explore the role of intestinal flora related to obesity after Ganpu tea enters the body,this experiment is explored through the following research:(1)The content of representative active components related to obesity in different varieties of Ganpu tea(Xiaoqing,Erhong and Dahong)was analyzed.After determination by HPLC,the contents of caffeine,gallic acid,tangerine peel and hesperidin in Xiaoqing were significantly higher than those in Erhong and Dahong.(2)C57 mice were used as model animals to explore the effects of different concentrations of Ganpu tea on the prevention of obesity in healthy mice under the condition of normal diet.After feeding for 3 weeks,the physiological indexes related to obesity were measured.Mice were weighed for body weight and relative weight of adipose tissue,and H&E-stained sections of adipose tissue and serum adiposity index were observed.Then,the gut microbiota structure and SCFAs production were studied by 16 SrRNA amplification and high-throughput sequencing.The results showed that Ganpu tea inhibited the weight growth of healthy mice and was beneficial to the adjustment of intestinal microbial structure,Low concentration of Ganpu tea increased the concentration of short chain fatty acids.(3)Under the condition of high-fat diet,the effects of Ganpu tea on various obesity phenotypes of dietary obese mice were studied.After 10 weeks of treatment with Ganpu tea,the body weight,adipose tissue,serum fat index,oxidative stress parameters and inflammatory factors of high-fat obese mice were measured,and the liver and adipose tissue were observed.The effects of Ganpu tea on gut microbiota and metabolism of dietary obese mice were analyzed by 16 SrRNA sequencing and fecal metabolome.The results showed that Ganpu tea could regulate the increase of Firmicutes abundance and the significant decrease of Lactobacillus caused by high-fat diet.Metabolome analysis showed that there were significant differences in obesity related marker metabolites such as 4-aminobutyral,d-Gluconic acid and zearalenol after citrus tea treatment. |
