| Health is the eternal pursuit of human beings.In vitro diagnosis(IVD),an important branch of the medical industry,has become an indispensable tool for disease prevention,clinical diagnosis and condition monitoring,including biochemical diagnosis,immunodiagnosis,molecular diagnosis,point-of-care diagnosis and so on.Polystyrene fluorescent microspheres is one of the core raw materials for in vitro diagnosis.Its preparation and performance optimization are critical for achieving accurate detection in various fields.So far,many methods have been developed by domestic and foreign scholars to prepare polystyrene fluorescent microspheres.The swelling method has attracted much attention because of its simple operation and strong practicability.It is necessary to further explore the factors related to the loading of fluorescent substances in the swelling process,which has great significance for the preparation of high-performance fluorescent microspheres and the realization of high sensitivity detection.Based on the above content,polystyrene fluorescent microspheres were prepared by swelling method.The research proceeds around microspheres′structure,diffusion process and application.The specific work is as follows:1 Preparation and characterization of solid polystyrene fluorescent microspheresWhen solid polystyrene fluorescent microspheres were prepared by swelling method,the internal chain structure of microspheres would affect the incorporation of dyes,and then affect the luminescence properties of the prepared microspheres.In this chapter,the soap-free emulsion polymerization was utilized to prepare solid microspheres with different chain structures by adjusting types of comonomers.Then,swelling was carried out.The experimental results showed that the swelling properties can be effectively improved by introducing lauryl methacrylate.When 10% lauryl methacrylate was added after 2 h of reaction,the microspheres had uniform particle size and good monodispersity.The loading amount of europium complex reached the maximum of 47.10 mg/g after swelling the prepared microspheres.2 Preparation and characterization of hollow polystyrene fluorescent microspheresIn order to explore the influence of spatial structure on the incorporation of dyes,polystyrene seed spheres was first synthesized in this chapter.The seed polymerization method was utilized to prepare hollow polymer microspheres with different spatial structures by adjusting types and amounts of shell monomers.Then,swelling was carried out.The experimental results showed that the microspheres had good uniformity and obvious cavity structure when the addition amount of hydroxyethyl methacrylate and divinylbenzene was 0.2 mL and 0.8 mL respectively.After swelling,the microspheres had high emission intensity and good stability.3 Study on the diffusion process of polystyrene fluorescent microspheres prepared by swelling method and its application in immunochromatographic detectionThe influence of diffusion process is preliminarily explored by control variable method in this chapter.In addition,the fluorescent microspheres with the best performance was selected for carboxyl groups content determination and protein adsorption performance evaluation,C-reactive protein immunoassay test strip was then prepared.The experimental results showed that the surface carboxyl groups content was0.467 mmol/g,and the adsorption capacity of BSA was 0.0137 mg/mg.The detection range of the prepared test strip was 1.3-210.7 mg/L,and the repeatability was desirable. |
