Ferulic oligosaccharides(FOs)are functional oligosaccharides formed by ferulic acid(FA)and oligosaccharides through ester bonds.Previous study found that FOs can predict and regulate diabetes by regulating advanced glycation end products.Therefore,in this paper,FOs were used as wall material components to study the oxygen barrier function of FOs in the wall material of microcapsules.The regulatory mechanism of FOs on redox level and metabolism was also explored from the aspects of simulated digestion in vitro and hepatointestinal metabolism in vivo.The purpose is to study the biological activity and function of FOs as food additives,and provide theoretical basis for the application of FOs in the field of microcapsule wall materials.The main research content and results of this paper are as follows:(1)First,FOs were prepared by fermentation and enzymatic hydrolysis,and their structures were identified to study the structural differences between the two FOs.The results showed that the composition of FOs by fermentation method was 738 mg/g of total sugar,121 mg/g of reducing sugar,27.43 mg/g of FA,and the main degree of polymerization was 4~6.The composition of FOs by enzymolysis method was 767mg/g of total sugar,142 mg/g of reducing sugar,41.25 mg/g of FA,and the main degree of polymerization was 4~8.Both monosaccharide components contain xylose and arabinose.The main difference between FOs prepared by biological fermentation and enzymatic hydrolysis is the content of FA and the degree of polymerization.(2)Secondly,FOs and OSA-starch were used as wall materials,β-Carotene was used as the core material to further study the oxygen barrier effect of FOs in microcapsule systems.The results showed that the shape of the microcapsules containing FOs in the wall material was relatively complete and nearly spherical.With the increase of the proportion of FOs in the wall material components,the membrane oxygen permeability of the wall material decreases,the storage stability of the core material increases,and the oxidation resistance of the microcapsule system increases.It shows that FOs have both oxygen blocking and oxygen quenching effects,and are a good composite component of wall materials.(3)Then the anti-digestion effect of FOs on starch was studied.The results showed that FOs affect the physicochemical properties of starch.At 10.0%,FOs can inhibit theα-Amylase activity reduces the content of quick-digestible starch in waxy corn starch.Tea polyphenols and ferulic acid can significantly affect the physicochemical properties of three kinds of corn starch and inhibit α-Amylase activity reduced the proportion of fast digestible starch,while xylo-oligosaccharide had no significant difference with the blank group.It shows that FOs only have anti-digestion effect on starch at high dosage and is the result of ferulic acid in structure.There is no significant anti-digestibility under daily intake conditions.(4)Finally,animal experiments were conducted to investigate the regulatory effects of FOs microcapsules on antioxidant and hepatointestinal metabolism in mice during metabolic processes.The results showed that under the condition of low dose(250 mg/kg/d),the antioxidant capacity of mouse serum and liver was increased,and the harmful bacteria in the intestine of mice fed with high-fat biscuits were inhibited at the level of phylum and genus,and the proportion of Firmicutes and Bacteroidota was adjusted(F: B).However,at medium and high doses(500,1250 mg/kg/d),the contents of malondialdehyde(MDA),total bile acid(TBA),total bilirubin(TBIL)and the activity of alkaline phosphatase(AKP)in mice increased significantly,and the liver of mice in high dose group showed pathological changes.It shows that low dose intake of FOs can increase the antioxidant capacity of the body and regulate metabolic levels.However,high dose intake will lead to the disorder of oxidation regulation level and intestinal flora,and lead to the disorder of bile secretion,leading to liver inflammation in mice.In summary,FOs are a good composite component of microcapsule wall materials,which have both oxygen barrier and quenching effects in the wall material system.Low dose intake can improve the antioxidant capacity and regulate metabolism in the body,but high dose intake may lead to bile secretion imbalance,leading to hepatitis.
