In the cosmetics industry,many products use anti-oxidant substances,such as dibutyl hydroxytoluene,vitamin C,and vitamin E.Although these substances have strong antioxidant activity,they have the weakness of poor stability.Therefore,it is the goal that people have been pursuing to find more antioxidant active substances with high stability and good safety,and to apply them to industrial production.Since our country has abundant marine biological resources,discovering new types of antioxidant active substances from the ocean has broad research space.This topic is to screen safe and stable antioxidant activity peptides from seaweed and apply them to the cosmetics industry as an important raw material,laying an experimental foundation for the application of new products.First of all,through in vitro experiments,the free radical scavenging rate and the anti-superoxide anion activity of the peptide solution were determined.The experimental results showed that among the 10 kinds of seaweed-derived peptide solutions,the peptide No.2 has the most stable antioxidant properties.At the same time,the formulation of the emulsion base is also designed.After the design of the raw material ratio,the single-factor experiment optimization is carried out,and then the response surface optimization design is carried out to screen and establish the best formula.The experimental results show that with carbomer as 0.05 g,emulsifier as2.53%,and rotating speed as 3.47 K/rim,the preparation parameters obtained are as follows: isopropyl palmitate 6%,stearic acid 2%,stearyl alcohol 1%,Span 20 1%,Tween 20 1.8%,glycerin 5%,propylene glycol 3%,carbomer 0.5%,triethanolamine0.5%,1,2-hexanediol 0.05%.Secondly,optimize the emulsion preparation process.Using viscosity,particle size and appearance as the inspection indicators,single-factor experiments were carried out on the preparation process to determine the optimization interval of the key factors,and the orthogonal experiment design was carried out by using computer software.The optimized experiment results show that the best preparation process for the active peptide emulsion matrix is: the emulsifier needs to be added to the two phases during the emulsification process;the water phase raw material is mixed into the oil phase raw material for stirring during the synthesis process,and the optimum mixing time is 6 min.Then,design the safety and corrosion resistance of the emulsion.Three preservatives were compounded to carry out the anti-corrosion challenge experiment,and the best anti-corrosion system was obtained: 0.1% p-hydroxyacetophenone,0.15% chlorophenylglycol,0.2% 1,2-hexanediol.The stability inspection was carried out again,and the result showed that it conformed to the industry standard test without deterioration and met the standard requirements.At the same time,a safety experiment was carried out through chicken embryo fluff to verify that after adding different contents of seaweed active peptides,the irritation IS of the antioxidant emulsion was determined to be less than 1,and there was no irritation.Finally,the anti-oxidant efficacy of the seaweed peptide emulsion in vivo was tested.In vivo experiments,through the establishment of a mouse photoaging model test,the results show that the emulsion containing No.2 peptide has a certain protective effect on the skin,which improves the activity of antioxidant enzymes in the skin and reduces the damage in the skin tissues.The content of malondialdehyde(MDA),long-term use,will help increase the content of hydroxyproline(HYP)in the tissues,thereby achieving the effect of slowing down skin aging.In summary,this experiment carried out formula design,process optimization,safety evaluation,and efficacy measurement of seaweed peptide emulsion,and provided a new anti-oxidant seaweed peptide skin care solution that can scavenge free radicals and slow down skin aging.
