| Broccoli and its sprouts are more and more favored and concerned by the market because of their rich nutrition and multiple therapeutic health care effects,and their rich unique substance sulforaphane(SF)helps prevent the occurrence of chronic diseases such as cancer and cardiovascular diseases.Previous studies have shown that broccoli sprouts are a good source of sulforaphane,but there is a high cost of sulforaphane acquisition,so it is feasible to increase the content of sulforaphane in broccoli sprouts at low cost.Plasma is an emerging processing technology in recent years.In this study,we will explore the effect of plasma treatment on sulforaphane enrichment in broccoli sprouts,optimize the treatment conditions,and further enrich sulforaphane and other bioactive substances by adding exogenous sulfur sources.The mechanism of action of plasma treatment affecting the production and accumulation of bioactive substances in broccoli sprouts was revealed through metabolic mechanisms and transcriptomics.Based on the above high enrichment of biologically active substance broccoli sprouts,a broccoli sprout compound fruit and vegetable juice with both flavor and nutrition was developed.In this way,it provides theoretical basis and method support for the industrial production of broccoli sprouts and the development of corresponding functional foods.The main contents and results of this study are as follows:1.To investigate the effects of different plasma treatment conditions on sulforaphane content in broccoli sprouts.The results showed that the plasma had the highest germination status and active substance content under the conditions of voltage 30 KV and treatment time of 2 min;Secondly,under the optimal conditions of plasma treatment,the content of biologically active substances in different varieties of broccoli sprouts was studied.It shows that the content of active substances in different varieties after plasma treatment shows differences.Among them,YF have the highest content of sulforaphane,glucosinolates glycosides and melanocyte enzyme activities.2.Plasma treatment of 30 KV 2 min has the highest content of biologically active substances for broccoli sprouts.On this basis,the non-target metabolomics and transcriptomics of broccoli sprouts after plasma treatment were investigated,and 6739different metabolites were screened,of which amino acids and their derivatives and organic acids were the most numerous;7460 differentially expressed genes,and each enrichment analyzed the 20 most significant differential pathways.The results show that plasma treatment can induce changes in the expression regulation of certain metabolites and genes of broccoli sprout active substances.3.The application of different sulfur-containing compounds and combined plasma treatments had a significant effect on the germination status and active substance content of broccoli seeds.Compared with the control group,plasma and sulfur-containing compounds alone and in combination with plasma significantly improved sulforaphane,thioglucidin,total phenol and total flavonoid content,as well as melanocyte mustard enzyme activity and total antioxidant activity.However,the combined treatment of the two only significantly increased the sulforaphane and total flavonoid content compared with the treatment of sulfur-containing compounds alone;Compared with the Zn SO4 and Mg SO4treatment groups,the seedlings treated with Zn SO4+Mg SO4 had higher sulforaphane and total phenol content.Overall,the combination of plasma treatment and exogenous spraying Zn SO4+Mg SO4 treatment of broccoli seeds is the most effective for the enrichment of bioactive substances such as sulforaphane and has the best antioxidant capacity.4.To discuss the effects of three(55°C bleaching,steam scalding and blanching)heat treatment methods and different treatment times on the biologically active substances and quality of sprout juice.The results show that the sprout juice is optimal in terms of flavor,color and active substance content after steam hot blanching treatment.The combination of sprout juice and orange juice after three heat treatment methods with optimal treatment time was selected for sensory evaluation.The results showed that the amount of addition was 40%and the sprout juice was mixed with orange juice after 4 minutes of steam hot blanching,and the effect was the best. |
